Associated Press June 15, 2009The U.S. National Park Service is looking to stimulate summer vacations at national parks, starting this weekend.Interior Secretary Ken Salazar announced earlier this month that entrance fees at 147 national parks and monuments—including the Grand Canyon and Yosemite—will be waived on three weekends this summer. The weekends are June 20 and 21, July 18 and 19, and August 15 and 16."During these tough economic times, our national parks provide opportunities for affordable vacations for families," Salazar said at a news conference at Cuyahoga Valley National Park in Ohio."I encourage everyone to visit one of our nation's crown jewels this summer and especially to take advantage of the three free-admission weekends."Most Americans live less than a day's drive from a national park, Salazar said. Last year national parks attracted more than 275 million visits, generating an estimated U.S. $10.6 billion for local economies and supporting more than 213,000 jobs, he said.For the Park Service, the free weekends will mean a loss of an estimated half million dollars a day from entrance fees that range from $3 to $25. A total of 147 parks and monuments charge entrance fees. The country's other 244 parks are already free.Kendra Barkoff, a spokesperson for Salazar, said the lost revenue should be more than offset by an increase in park tourism. Many tour operators, hotels, restaurants, gift shops, and other vendors near national parks will offer other discounts and special promotions on the free-weekend dates, she said.The waiver applies only to entrance fees and does not affect charges for camping, reservations, tours, or concessions, Salazar said.Senator Max Baucus, a Democrat representing Montana, applauded the free weekends. Baucus has co-sponsored a bill that would cap park entrance fees at current rates unless approved by Congress. The bill also would limit fees on national forests and other federally managed lands."There is nothing better than spending a weekend in Glacier or Yellowstone, and to be able to do it without straining the budget is even better," Baucus said in a statement. You wonder if you have made a wrong turn. All at once an immense gorge a mile deep and up to 18 miles wide opens up. The scale is so vast that even from the best vantage point only a fraction of the canyon's 277 miles can be seen.Nearly five million people travel here each year; 90 percent first see the canyon from the South Rim with its dramatic views into the deep inner gorge of the Colorado River. So many feet have stepped cautiously to the edge of major overlooks that in places the rock has been polished smooth. But most of the park's 1,904 square miles are maintained as wilderness. You can avoid crowds by hiking the park's many trails or driving to the cool evergreen forests of the North Rim where people are fewer and viewing is more leisurely. The Grand Canyon boasts some of the nation's cleanest air, with visibility averaging 90 to 110 miles. Increasingly, though, air pollution blurs vistas that once were sharp and rich hued. Hazy days have become more common, with visibility dropping as low as 40 miles. park Accurate MS-based Rab10 Phosphorylation Stoichiometry Determination as Readout for LRRK2 Activity in Parkinson's Disease [Research] By www.mcponline.org Published On :: 2020-09-01T00:05:24-07:00 Pathogenic mutations in the Leucine-rich repeat kinase 2 (LRRK2) are the predominant genetic cause of Parkinson's disease (PD). They increase its activity, resulting in augmented Rab10-Thr73 phosphorylation and conversely, LRRK2 inhibition decreases pRab10 levels. Currently, there is no assay to quantify pRab10 levels for drug target engagement or patient stratification. To meet this challenge, we developed an high accuracy and sensitivity targeted mass spectrometry (MS)-based assay for determining Rab10-Thr73 phosphorylation stoichiometry in human samples. It uses synthetic stable isotope-labeled (SIL) analogues for both phosphorylated and nonphosphorylated tryptic peptides surrounding Rab10-Thr73 to directly derive the percentage of Rab10 phosphorylation from attomole amounts of the endogenous phosphopeptide. The SIL and the endogenous phosphopeptides are separately admitted into an Orbitrap analyzer with the appropriate injection times. We test the reproducibility of our assay by determining Rab10-Thr73 phosphorylation stoichiometry in neutrophils of LRRK2 mutation carriers before and after LRRK2 inhibition. Compared with healthy controls, the PD predisposing mutation carriers LRRK2 G2019S and VPS35 D620N display 1.9-fold and 3.7-fold increased pRab10 levels, respectively. Our generic MS-based assay further establishes the relevance of pRab10 as a prognostic PD marker and is a powerful tool for determining LRRK2 inhibitor efficacy and for stratifying PD patients for LRRK2 inhibitor treatment. The sensory properties of sparkling wine depend on a complex interplay between the chemical and biochemical components in the final product. Glycoproteins have been linked to positive and negative qualities in sparkling wine, but the glycosylation profiles of sparkling wine have not been previously investigated in detail. We analysed the glyco/proteome of sparkling wines using protein- and glycopeptide-centric approaches. We developed an automated workflow that created ion libraries to analyse Sequential Window Acquisition of all THeoretical mass spectra (SWATH) Data Independent Acquisition (DIA) mass spectrometry data based on glycopeptides identified by Byonic. We applied our workflow to three pairs of experimental sparkling wines to assess the effects of aging on lees and of different yeast strains used in the Liqueur de Tirage for secondary fermentation. We found that aging a cuvée on lees for 24 months compared to 8 months led to a dramatic decrease in overall protein abundance and an enrichment in large glycans at specific sites in some proteins. Secondary fermentation of a Riesling wine with Saccharomyces cerevisiae yeast strain Siha4 produced more yeast proteins and glycoproteins than with S. cerevisiae yeast strain DV10. The abundance and glycosylation profiles of grape glycoproteins were also different between grape varieties. This work represents the first in-depth study into protein- and peptide-specific glycosylation in sparkling wines and describes a quantitative glycoproteomic SWATH/DIA workflow that is broadly applicable to other sample types.
