Consistent structure estimation of exponential-family random graph models with block structure

Abstract:
We consider the challenging problem of statistical inference for exponential-family random graph models based on a single observation of a random graph with complex dependence. To facilitate statistical inference, we consider random graphs with additional structure in the form of block structure. We have shown elsewhere that when the block structure is known, it facilitates consistency results for $M$-estimators of canonical and curved exponential-family random graph models with complex dependence, such as transitivity. In practice, the block structure is known in some applications (e.g., multilevel networks), but is unknown in others. When the block structure is unknown, the first and foremost question is whether it can be recovered with high probability based on a single observation of a random graph with complex dependence. The main consistency results of the paper show that it is possible to do so under weak dependence and smoothness conditions. These results confirm that exponential-family random graph models with block structure constitute a promising direction of statistical network analysis.

Recurrence of multidimensional persistent random walks. Fourier and series criteria

Abstract:
The recurrence and transience of persistent random walks built from variable length Markov chains are investigated. It turns out that these stochastic processes can be seen as Lévy walks for which the persistence times depend on some internal Markov chain: they admit Markov random walk skeletons. A recurrence versus transience dichotomy is highlighted. Assuming the positive recurrence of the driving chain, a sufficient Fourier criterion for the recurrence, close to the usual Chung–Fuchs one, is given and a series criterion is derived. The key tool is the Nagaev–Guivarc’h method. Finally, we focus on particular two-dimensional persistent random walks, including directionally reinforced random walks, for which necessary and sufficient Fourier and series criteria are obtained. Inspired by ( Adv. Math. 208 (2007) 680–698), we produce a genuine counterexample to the conjecture of ( Adv. Math. 117 (1996) 239–252). As for the one-dimensional case studied in ( J. Theoret. Probab. 31 (2018) 232–243), it is easier for a persistent random walk than its skeleton to be recurrent. However, such examples are much more difficult to exhibit in the higher dimensional context. These results are based on a surprisingly novel – to our knowledge – upper bound for the Lévy concentration function associated with symmetric distributions.

Normal approximation for sums of weighted $U$-statistics – application to Kolmogorov bounds in random subgraph counting

Abstract:
We derive normal approximation bounds in the Kolmogorov distance for sums of discrete multiple integrals and weighted $U$-statistics made of independent Bernoulli random variables. Such bounds are applied to normal approximation for the renormalized subgraph counts in the Erdős–Rényi random graph. This approach completely solves a long-standing conjecture in the general setting of arbitrary graph counting, while recovering recent results obtained for triangles and improving other bounds in the Wasserstein distance.

Operator-scaling Gaussian random fields via aggregation

Abstract:
We propose an aggregated random-field model, and investigate the scaling limits of the aggregated partial-sum random fields. In this model, each copy in the aggregation is a $pm 1$-valued random field built from two correlated one-dimensional random walks, the law of each determined by a random persistence parameter. A flexible joint distribution of the two parameters is introduced, and given the parameters the two correlated random walks are conditionally independent. For the aggregated random field, when the persistence parameters are independent, the scaling limit is a fractional Brownian sheet. When the persistence parameters are tail-dependent, characterized in the framework of multivariate regular variation, the scaling limit is more delicate, and in particular depends on the growth rates of the underlying rectangular region along two directions: at different rates different operator-scaling Gaussian random fields appear as the region area tends to infinity. In particular, at the so-called critical speed, a large family of Gaussian random fields with long-range dependence arise in the limit. We also identify four different regimes at non-critical speed where fractional Brownian sheets arise in the limit.

Cliques in rank-1 random graphs: The role of inhomogeneity

Abstract:
We study the asymptotic behavior of the clique number in rank-1 inhomogeneous random graphs, where edge probabilities between vertices are roughly proportional to the product of their vertex weights. We show that the clique number is concentrated on at most two consecutive integers, for which we provide an expression. Interestingly, the order of the clique number is primarily determined by the overall edge density, with the inhomogeneity only affecting multiplicative constants or adding at most a $log log (n)$ multiplicative factor. For sparse enough graphs the clique number is always bounded and the effect of inhomogeneity completely vanishes.

A new method for obtaining sharp compound Poisson approximation error estimates for sums of locally dependent random variables

Abstract:
Let X 1 , X 2 , …, X n be a sequence of independent or locally dependent random variables taking values in ℤ + . In this paper, we derive sharp bounds, via a new probabilistic method, for the total variation distance between the distribution of the sum ∑ i =1 n X i and an appropriate Poisson or compound Poisson distribution. These bounds include a factor which depends on the smoothness of the approximating Poisson or compound Poisson distribution. This “smoothness factor” is of order O( σ −2 ), according to a heuristic argument, where σ 2 denotes the variance of the approximating distribution. In this way, we offer sharp error estimates for a large range of values of the parameters. Finally, specific examples concerning appearances of rare runs in sequences of Bernoulli trials are presented by way of illustration.

Abstract:
Gaussian graphical models are useful tools for exploring network structures in multivariate normal data. In this paper we are interested in situations where data show departures from Gaussianity, therefore requiring alternative modeling distributions. The multivariate $t$ -distribution, obtained by dividing each component of the data vector by a gamma random variable, is a straightforward generalization to accommodate deviations from normality such as heavy tails. Since different groups of variables may be contaminated to a different extent, Finegold and Drton (2014) introduced the Dirichlet $t$ -distribution, where the divisors are clustered using a Dirichlet process. In this work, we consider a more general class of nonparametric distributions as the prior on the divisor terms, namely the class of normalized completely random measures (NormCRMs). To improve the effectiveness of the clustering, we propose modeling the dependence among the divisors through a nonparametric hierarchical structure, which allows for the sharing of parameters across the samples in the data set. This desirable feature enables us to cluster together different components of multivariate data in a parsimonious way. We demonstrate through simulations that this approach provides accurate graphical model inference, and apply it to a case study examining the dependence structure in radiomics data derived from The Cancer Imaging Atlas.

Noncoding Microdeletion in Mouse Hgf Disrupts Neural Crest Migration into the Stria Vascularis, Reduces the Endocochlear Potential, and Suggests the Neuropathology for Human Nonsyndromic Deafness DFNB39

Hepatocyte growth factor (HGF) is a multifunctional protein that signals through the MET receptor. HGF stimulates cell proliferation, cell dispersion, neuronal survival, and wound healing. In the inner ear, levels of HGF must be fine-tuned for normal hearing. In mice, a deficiency of HGF expression limited to the auditory system, or an overexpression of HGF, causes neurosensory deafness. In humans, noncoding variants in HGF are associated with nonsyndromic deafness DFNB39. However, the mechanism by which these noncoding variants causes deafness was unknown. Here, we reveal the cause of this deafness using a mouse model engineered with a noncoding intronic 10 bp deletion (del10) in Hgf. Male and female mice homozygous for del10 exhibit moderate-to-profound hearing loss at 4 weeks of age as measured by tone burst auditory brainstem responses. The wild type (WT) 80 mV endocochlear potential was significantly reduced in homozygous del10 mice compared with WT littermates. In normal cochlea, endocochlear potentials are dependent on ion homeostasis mediated by the stria vascularis (SV). Previous studies showed that developmental incorporation of neural crest cells into the SV depends on signaling from HGF/MET. We show by immunohistochemistry that, in del10 homozygotes, neural crest cells fail to infiltrate the developing SV intermediate layer. Phenotyping and RNAseq analyses reveal no other significant abnormalities in other tissues. We conclude that, in the inner ear, the noncoding del10 mutation in Hgf leads to developmental defects of the SV and consequently dysfunctional ion homeostasis and a reduction in the EP, recapitulating human DFNB39 nonsyndromic deafness.

SIGNIFICANCE STATEMENT Hereditary deafness is a common, clinically and genetically heterogeneous neurosensory disorder. Previously, we reported that human deafness DFNB39 is associated with noncoding variants in the 3'UTR of a short isoform of HGF encoding hepatocyte growth factor. For normal hearing, HGF levels must be fine-tuned as an excess or deficiency of HGF cause deafness in mouse. Using a Hgf mutant mouse with a small 10 bp deletion recapitulating a human DFNB39 noncoding variant, we demonstrate that neural crest cells fail to migrate into the stria vascularis intermediate layer, resulting in a significantly reduced endocochlear potential, the driving force for sound transduction by inner ear hair cells. HGF-associated deafness is a neurocristopathy but, unlike many other neurocristopathies, it is not syndromic.

Rapid Release of Ca2+ from Endoplasmic Reticulum Mediated by Na+/Ca2+ Exchange

Phototransduction in Drosophila is mediated by phospholipase C (PLC) and Ca2+-permeable TRP channels, but the function of endoplasmic reticulum (ER) Ca2+ stores in this important model for Ca2+ signaling remains obscure. We therefore expressed a low affinity Ca2+ indicator (ER-GCaMP6-150) in the ER, and measured its fluorescence both in dissociated ommatidia and in vivo from intact flies of both sexes. Blue excitation light induced a rapid (tau ~0.8 s), PLC-dependent decrease in fluorescence, representing depletion of ER Ca2+ stores, followed by a slower decay, typically reaching ~50% of initial dark-adapted levels, with significant depletion occurring under natural levels of illumination. The ER stores refilled in the dark within 100–200 s. Both rapid and slow store depletion were largely unaffected in InsP3 receptor mutants, but were much reduced in trp mutants. Strikingly, rapid (but not slow) depletion of ER stores was blocked by removing external Na+ and in mutants of the Na+/Ca2+ exchanger, CalX, which we immuno-localized to ER membranes in addition to its established localization in the plasma membrane. Conversely, overexpression of calx greatly enhanced rapid depletion. These results indicate that rapid store depletion is mediated by Na+/Ca2+ exchange across the ER membrane induced by Na+ influx via the light-sensitive channels. Although too slow to be involved in channel activation, this Na+/Ca2+ exchange-dependent release explains the decades-old observation of a light-induced rise in cytosolic Ca2+ in photoreceptors exposed to Ca2+-free solutions.

SIGNIFICANCE STATEMENT Phototransduction in Drosophila is mediated by phospholipase C, which activates TRP cation channels by an unknown mechanism. Despite much speculation, it is unknown whether endoplasmic reticulum (ER) Ca2+ stores play any role. We therefore engineered flies expressing a genetically encoded Ca2+ indicator in the photoreceptor ER. Although NCX Na+/Ca2+ exchangers are classically believed to operate only at the plasma membrane, we demonstrate a rapid light-induced depletion of ER Ca2+ stores mediated by Na+/Ca2+ exchange across the ER membrane. This NCX-dependent release was too slow to be involved in channel activation, but explains the decades-old observation of a light-induced rise in cytosolic Ca2+ in photoreceptors bathed in Ca2+-free solutions.

Young Catholics in Indonesia provide aid amid coronavirus

CNA Staff, Apr 23, 2020 / 05:13 pm (CNA).- A Catholic youth organization in Indonesia has instituted a movement to provide assistance to families struggling during the coronavirus pandemic.

Orang Muda Katolik, or Catholic Young People, recently began the initiative “Adopt One Brother,” which encourages youth to volunteer time and money to support poorer families, many of whom are now unemployed.

Indonesia has over 7,500 cases of COVID-19, and 647 deaths. According to data from the country’s Ministry of Labour, Aljazeera reported, 2.8 million Indonesians have lost their jobs because of the pandemic.

Stefanus Gusma, who leads OMK’s COVID-19 task force, said the initiative has spread to 26 of the country’s 34 provinces and involved thousands of OMK members. He said volunteers are encouraged to donate 200,000 to 500,000 rupiah ($12-32) per week.

"First, we mobilized our own members to help our fellow brothers and sisters who are experiencing difficulties. Then we extended our reach to anyone who was willing to help others,” Gusma told UCA News.

"After we receive their data, we contact them about where they would like their donations to go,” he said. “If a donor wants to donate to a family in East Nusa Tenggara province, we will coordinate with our members there to seek a family in need.”

With help from the local dioceses and governments, the organization has also distributed about 2,000 aid packages, electricity vouchers, and hygienic products.

According to UCA News, other OMK members said the organization has not only provided aid to families but to hospitals and orphanages as well. Maskendari, an OMK member in Pontianak, said the organization has distributed “hundreds of aid packages and thousands of personal protection items such as masks and bottles of hand sanitizer.”

“We want others to act, not only through our organization but also individually or with other groups,” Gusma told UCA News. "We want to show the importance of showing human solidarity in the midst of this current crisis," he added.

Orang Muda Katolik seeks to mentor young Catholics, aged between 15 and 35, by providing educational resources, coaching, and volunteer opportunities.

Bishop Pius Prapdi of Ketapang issued a letter to OMK at the end of March. He encouraged young Catholics to follow social distancing rules and other safety precautions. However, he also challenged the youth to find creative ways to help the community, like investigating free food assistance for those in need and checking-in on neighbors through social media.

“Catholic Young People can also help others in a safe way,” he wrote. “With creativity, young people can become leaders in this situation and go through critical times together.”

“Pope Francis invites young people to become the main actors (protagonists) in renewing the world, let us in this crisis period stop for a moment to reflect back on what we have made for ourselves, the environment, the Church and the citizens of the world.”

Pandemic brings ‘a very different kind of Church’ to London’s homeless

London, England, May 5, 2020 / 09:00 am (CNA).- A parish in London’s West End is offering the homeless adoration, access to sacraments, and the rosary -- along with food provided by a five-star restaurant. 

St. Patrick’s Church in Soho, an area known for its nightlife and red-light district, is offering a remarkable ministry to the homeless as the capital struggles to cope with the coronavirus pandemic. 

Pastor Fr. Alexander Sherbrooke said he had “a strong sense that the Holy Spirit is literally building a church on the streets” in response to the crisis. 

When the city began to shut down in mid-March, Westminster City Council turned to Sherbrooke, who has overseen daily outreach to the homeless since he arrived in the parish in 2001.

He told CNA that the council had asked St. Patrick’s to increase its provision of food to the homeless significantly while it tried to house those living on the streets. 

The parish, founded in 1792, had previously fed the homeless in its parish center. But after Catholic churches across the country were ordered to close because of the virus, St. Patrick’s was forced to improvise. It began serving the homeless food on its doorstep twice a day, Monday through Sunday.

“On most days we are providing up to 320 meals,” Sherbrooke explained. “On average, we probably see 220 people a day, some of whom come for both breakfast and dinner.”

Hot food is supplied by the Connaught Hotel, a five-star restaurant in London’s affluent Mayfair district, as well as by Wiltons Restaurant in Jermyn Street. The Pret a Manger chain provides sandwiches. 

“It’s a very sophisticated operation and we fully intend to be diligent in preserving social distancing, personal hygiene, food hygiene, etc,” the priest said. “We have a good number of volunteers. We also continue to provide a shower and lavatory facility.” 

Sherbrooke explained that the homeless in the West End live off the footfall generated by local businesses, restaurants and theaters. 

“There is none of that now,” he said. “It’s amazingly empty and can be quite intimidating, particularly at nighttime.”

”The West End has many who are alcohol and drug dependent and without their normal source of income, this can create a volatile situation. Police are very present, but the West End is very inhospitable, at times threatening and not very pleasant.”

“I’ve been in the parish for some 17 years, throughout which  much of my time has been spent in pastoral care for those who are needy. But nothing has really prepared me for where we are at the moment.”

Volunteers at St. Patrick’s are determined to relieve spiritual as well as physical deprivation. As food is distributed, they pray before the Blessed Sacrament in a nearby adoration tent, while observing social distancing. Sherbrooke is available for visitors seeking a sacramental encounter, sitting at a safe distance and behind a white sheet. There is also a tent offering lectio divina. 

“This enhanced feeding facility has come very much as a response to the request of the local authority,” Sherbrooke said. “We have a long tradition of feeding people happily and well. But in a very strange sort of way, the Church, from being a physical reality behind four walls, is now a reality in the street.”

Sherbrooke, who cites St Damien of Molokai and Mother Teresa as inspirations, continued: “It’s imparting a spiritual, pastoral care, where I have a strong sense that the Holy Spirit is literally building a church on the streets. There’s lectio divina. There’s adoration -- in other words, a prolongation of the Holy Mass -- confession, rosary, etc.”

“We are ministering to the people. We are going to them, speaking to them, giving rosaries and sharing the Gospel. So there is a real work of evangelization going on.”

Volunteers also distribute a sheet each week with reflections, Scripture readings, and advice on how to pray.

“So there’s a kind of catechesis of the poor which is going on,” Sherbrooke said.

“There is a very real sense that in this terrible virus situation that God is creating a very different kind of Church, much more evangelical, and perhaps simpler. All this has happened not through management but I believe through God's providence.”

He noted that despite the present dangers volunteers felt a strong sense of supernatural protection. 

“Personally, I would say that the way that I haven’t caught [the virus] -- given the reality of the situation here -- is that every day I pray that the Precious Blood of Jesus will come into my heart, my veins, my lungs, and protect me from the virus so that I can do this work,” he said. 

In 2011, St. Patrick’s reopened after a £4 million restoration project, which included the excavation of the basement and the creation of the parish center, located beneath the church. Food for the homeless is now prepared there every day.

“It’s almost as though God has crafted this parish for this work at this time,” Sherbrooke said.

Noninvasive Ventilation for Respiratory Distress Syndrome: A Randomized Controlled Trial

Nasal continuous positive airway pressure (NCPAP) has been the initial respiratory support for many preterm infants with respiratory distress syndrome (RDS). Nasal intermittent positive-pressure ventilation (NIPPV) seems to increase the beneficial effects of NCPAP by combining it with ventilatory inflations.

This study suggests that NIPPV, as an intial respiratory support for preterm infants with RDS, is feasible and safe and may have beneficial effects, when compared with NCPAP. (Read the full article)

Corticosteroid Pulse Combination Therapy for Refractory Kawasaki Disease: A Randomized Trial

The efficacy of intravenous immunoglobulin and corticosteroid pulse combination therapy for refractory Kawasaki disease has been established. The Egami score can be used to predict which patients are likely to have refractory Kawasaki disease.

As a new strategy for primary treatment, intravenous immunoglobulin and corticosteroid pulse combination therapy is safe and effective for patients predicted to have refractory Kawasaki disease based on the Egami score. (Read the full article)

Peer-led Education for Adolescents With Asthma in Jordan: A Cluster-Randomized Controlled Trial

The prevalence of asthma and smoking among adolescents in Jordan is high. Well-designed, school-based, peer-led education programs can have a positive impact on asthma self-management in adolescents. Student peer leaders can be useful and responsible partners in health promotion programs.

A peer-led asthma education program —Adolescent Asthma Action—for adolescents developed in Australia was adapted to suit non–English-speaking cultures in the Middle East. Early Intervention Improves Behavioral Outcomes for Preterm Infants: Randomized Controlled Trial

Prematurely born children have an increased prevalence of behavioral problems in the long term. Knowledge regarding the effects of early intervention programs is sparse, and more randomized controlled trials are warranted.

A modified version of the Mother-Infant Transaction Program showed beneficial effects on behavioral outcomes reported by both parents of preterm infants with birth weights of <2000 g at a corrected age of 5 years. (Read the full article)
