NHL calls off 2020-21 season games in Helsinki, Prague

National Hockey League regular-season games that were scheduled for early in the 2020-21 campaign in Helsinki and Prague have been called off in the wake of the coronavirus pandemic. With concern about when or if the halted 2019-20 campaign might resume due to the deadly virus outbreak, the NHL and the NHL Players Association announced the decision to scrap the matchups with an eye to a 2021 return. "The NHLPA and the NHL remain committed to maintaining and growing our international presence," a joint statement said.

High-dimensional Gaussian graphical models on network-linked data

Graphical models are commonly used to represent conditional dependence relationships between variables. There are multiple methods available for exploring them from high-dimensional data, but almost all of them rely on the assumption that the observations are independent and identically distributed. At the same time, observations connected by a network are becoming increasingly common, and tend to violate these assumptions. Here we develop a Gaussian graphical model for observations connected by a network with potentially different mean vectors, varying smoothly over the network. We propose an efficient estimation algorithm and demonstrate its effectiveness on both simulated and real data, obtaining meaningful and interpretable results on a statistics coauthorship network. We also prove that our method estimates both the inverse covariance matrix and the corresponding graph structure correctly under the assumption of network “cohesion”, which refers to the empirically observed phenomenon of network neighbors sharing similar traits.

Abstract:
In actuarial research a task of particular interest and importance is to predict the loss cost for individual risks so that informative decisions are made in various insurance operations such as underwriting, ratemaking and capital management. The loss cost is typically viewed to follow a compound distribution where the summation of the severity variables is stopped by the frequency variable. A challenging issue in modeling such outcomes is to accommodate the potential dependence between the number of claims and the size of each individual claim. In this article we introduce a novel regression framework for compound distributions that uses a copula to accommodate the association between the frequency and the severity variables and, thus, allows for arbitrary dependence between the two components. We further show that the new model is very flexible and is easily modified to account for incomplete data due to censoring or truncation. The flexibility of the proposed model is illustrated using both simulated and real data sets. In the analysis of granular claims data from property insurance, we find substantive negative relationship between the number and the size of insurance claims. In addition, we demonstrate that ignoring the frequency-severity association could lead to biased decision-making in insurance operations.

Incorporating conditional dependence in latent class models for probabilistic record linkage: Does it matter?

Abstract:
The conditional independence assumption of the Felligi and Sunter (FS) model in probabilistic record linkage is often violated when matching real-world data. Ignoring conditional dependence has been shown to seriously bias parameter estimates. However, in record linkage, the ultimate goal is to inform the match status of record pairs and therefore, record linkage algorithms should be evaluated in terms of matching accuracy. In the literature, more flexible models have been proposed to relax the conditional independence assumption, but few studies have assessed whether such accommodations improve matching accuracy. In this paper, we show that incorporating the conditional dependence appropriately yields comparable or improved matching accuracy than the FS model using three real-world data linkage examples. Through a simulation study, we further investigate when conditional dependence models provide improved matching accuracy. Our study shows that the FS model is generally robust to the conditional independence assumption and provides comparable matching accuracy as the more complex conditional dependence models. However, when the match prevalence approaches 0% or 100% and conditional dependence exists in the dominating class, it is necessary to address conditional dependence as the FS model produces suboptimal matching accuracy. The need to address conditional dependence becomes less important when highly discriminating fields are used. Our simulation study also shows that conditional dependence models with misspecified dependence structure could produce less accurate record matching than the FS model and therefore we caution against the blind use of conditional dependence models.

3 NY children die from syndrome possibly linked to COVID-19

Three children have now died in New York state from a possible complication from the coronavirus involving swollen blood vessels and heart problems, Gov. Andrew Cuomo said Saturday. At least 73 children in New York have been diagnosed with symptoms similar to Kawasaki disease — a rare inflammatory condition in children — and toxic shock syndrome.

Molecular Mechanisms of Non-ionotropic NMDA Receptor Signaling in Dendritic Spine Shrinkage

Structural plasticity of dendritic spines is a key component of the refinement of synaptic connections during learning. Recent studies highlight a novel role for the NMDA receptor (NMDAR), independent of ion flow, in driving spine shrinkage and LTD. Yet little is known about the molecular mechanisms that link conformational changes in the NMDAR to changes in spine size and synaptic strength. Here, using two-photon glutamate uncaging to induce plasticity at individual dendritic spines on hippocampal CA1 neurons from mice and rats of both sexes, we demonstrate that p38 MAPK is generally required downstream of non-ionotropic NMDAR signaling to drive both spine shrinkage and LTD. In a series of pharmacological and molecular genetic experiments, we identify key components of the non-ionotropic NMDAR signaling pathway driving dendritic spine shrinkage, including the interaction between NOS1AP (nitric oxide synthase 1 adaptor protein) and neuronal nitric oxide synthase (nNOS), nNOS enzymatic activity, activation of MK2 (MAPK-activated protein kinase 2) and cofilin, and signaling through CaMKII. Our results represent a large step forward in delineating the molecular mechanisms of non-ionotropic NMDAR signaling that can drive shrinkage and elimination of dendritic spines during synaptic plasticity.

SIGNIFICANCE STATEMENT Signaling through the NMDA receptor (NMDAR) is vitally important for the synaptic plasticity that underlies learning. Recent studies highlight a novel role for the NMDAR, independent of ion flow, in driving synaptic weakening and dendritic spine shrinkage during synaptic plasticity. Here, we delineate several key components of the molecular pathway that links conformational signaling through the NMDAR to dendritic spine shrinkage during synaptic plasticity.

Nestin Selectively Facilitates the Phosphorylation of the Lissencephaly-Linked Protein Doublecortin (DCX) by cdk5/p35 to Regulate Growth Cone Morphology and Sema3a Sensitivity in Developing Neurons

Nestin, an intermediate filament protein widely used as a marker of neural progenitors, was recently found to be expressed transiently in developing cortical neurons in culture and in developing mouse cortex. In young cortical cultures, nestin regulates axonal growth cone morphology. In addition, nestin, which is known to bind the neuronal cdk5/p35 kinase, affects responses to axon guidance cues upstream of cdk5, specifically, to Sema3a. Changes in growth cone morphology require rearrangements of cytoskeletal networks, and changes in microtubules and actin filaments are well studied. In contrast, the roles of intermediate filament proteins in this process are poorly understood, even in cultured neurons. Here, we investigate the molecular mechanism by which nestin affects growth cone morphology and Sema3a sensitivity. We find that nestin selectively facilitates the phosphorylation of the lissencephaly-linked protein doublecortin (DCX) by cdk5/p35, but the phosphorylation of other cdk5 substrates is not affected by nestin. We uncover that this substrate selectivity is based on the ability of nestin to interact with DCX, but not with other cdk5 substrates. Nestin thus creates a selective scaffold for DCX with activated cdk5/p35. Last, we use cortical cultures derived from Dcx KO mice to show that the effects of nestin on growth cone morphology and on Sema3a sensitivity are DCX-dependent, thus suggesting a functional role for the DCX-nestin complex in neurons. We propose that nestin changes growth cone behavior by regulating the intracellular kinase signaling environment in developing neurons. The sex of animal subjects is unknown.

Thinking a Wrong Is Right

In 1973, after the United States Supreme Court legalized abortion in America, my dad—a doctor—was interviewed by the local paper about the ruling. One of his quotes became the story’s headline: “Just because it’s legal, doesn’t mean it’s right.”

I’ve never forgotten those words. Even as a second grader, they left a deep impression on me. I was only 8 years old, but I understood that no law could make what’s wrong right. No law could take away the dignity of the human person or make it okay to kill an unborn child.

Unfortunately, what I didn’t realize at the time is that while laws can’t make a wrong right, they can make people think a wrong is right. The law is teacher, and the law Roe v. Wade established has taught three generations of Americans that human persons are disposable. Along with the rest of what St. John Paul II called “the culture of death,” that ruling has tricked millions into believing that we can get rid of human beings when they inconvenience us or burden us.

This attitude puts countless lives in danger—not just the unborn, but also the elderly, the sick, the disabled, the poor, and the stranger. It also puts our entire culture in danger.

Choosing to love and care for the most vulnerable among us is not about politics. It’s not a prudential decision upon which people of good will can disagree. It is a moral imperative. Every other moral issue is related to recognizing the dignity of all human life. From the understanding that life is sacred and the human person is made in God’s image, every other action we call “good” flows.

Because of that, a culture that rejects the sacredness of life cannot endure. Everything that makes a culture healthy—honesty, trust, friendship, charity, kindness, courage—all of that hinges on the dignity of the human person. Take that away, and the rest will crumble. So will we.

Each of us faces the choice my father articulated back in 1973. Will we stand up for what is right, even when a law says we’re wrong? Or will we allow an unjust law to dictate what we believe and do?

On January 24, I will join hundreds of thousands of other Americans who are choosing to defend what is right, by participating in the 47th annual March for Life in Washington, D.C.

Every year, Franciscan University of Steubenville, which is both my alma mater and the school I serve as president, transports hundreds of students to the march. Together, we walk. Not because we expect one lone march to change things. But rather as a reminder to our culture that this isn’t an issue that will just go away.

No law legalizing abortion has settled the question. No law legalizing abortion ever will settle the question. Abortion is wrong, and people who recognize that are going to keep showing up and keep speaking up until the law recognizes that, too. Again, the law is a teacher, and our future as a nation depends upon it teaching what is right and true.

Despite what the media wants us to think, abortion is not a private matter. It wounds the women who believe they don’t have any other option. It wounds the families who lose babies to love. It wounds the health care workers, who buy into the lie of abortion. And it wounds our entire culture, choking the life out of it at its very roots.

The public devastation of abortion demands a public response. Yes, we must pray to end abortion. We must do everything we can to empower women to raise their children or place them in loving homes through adoption. But we also must continue to speak up. We must refuse to allow our faith in the dignity of human life to be pushed aside and kept out of public view. We must continue to march. When we do, we put the conscience of America on display. We remind people of what my dad always knew: Just because it’s legal, doesn’t mean it’s right.
