Bird watching festival underway

The Wetland Park is holding its annual Bird Watching Festival, with "Incredible Bird Parents" as this year’s theme.

 

Running until April 20, the event shows how versatile and sophisticated birds are in providing parental care for their offspring.

 

For the first time, five unique Lego brick wetland animal models are displayed as another festival attraction.

 

They include the saltwater crocodile Pui Pui, a kingfisher, paddy frog, fiddler crab and the common tiger, as well as Lego brick wall art.

 

Co-created by the Wetland Park and Lego Certified Professional Andy Hung, the Lego series aims to inspire kids to build a better future for nature.

 

Other edutainment comprises bird watching tours, thematic bird interpretation sessions, bird photography workshops, public lectures and a kids reading club.

Driving test services to resume

The Transport Department today announced that its driving test (road test) service will resume on May 4 to align with the resumption of public services in phases.

 

The department said candidates scheduled to take a road test on or after May 4 should take the test according to the test date, time and the driving test centre specified in their appointment letter.

 

Candidates affected by the suspension of driving test centres will be notified by post of the rescheduled test.

 

Driving test (written test Part A) and taxi written test services will resume on May 26.

 

Candidates affected by the suspension of written test services will be rescheduled to take the tests from May 26 and thereafter and notified by post of the rescheduling.

 

All candidates participating in the driving, road and written tests, must wear surgical masks inside the test centres and throughout the test.

 

They must also clean their hands with alcohol sanitisers and undergo body temperature screening.

 

If a candidate does not wear a surgical mask, refuses body temperature screening or has a body temperature higher than 37.5 degrees Celsius, the candidate will not be allowed to enter the venue and the test will be rescheduled.

 

To improve air circulation inside the vehicle compartment, driving examiners may open vehicle windows during the test.

 

The department urged candidates not to attend a driving test if they are unwell.

 

For applicants who are absent from a driving test on medical grounds, they may submit their postponement applications with the original sick leave or medical certificate to the Driving Test Appointment Office within one month from the test date for rescheduling.

 

Candidates applying for a postponement, temporary cancellation of driving test appointment or change of driving test region may submit signed application letters, copies of identification document and driving test appointment letter via the drop-in box at the Kowloon Licensing Office or post them to the Driving Test Appointment Office on 2/F, Cheung Sha Wan Government Offices, 303 Cheung Sha Wan Road.

 

Application of driving test appointment services for fresh candidates and repeaters will continue to be suspended until further notice. Call 2771 7723 for enquiries.

 

Vehicle Examination Centres will resume full services on May 4. The department will continue to process applications for licensing and related services submitted by applicants with scheduled appointments via the drop-in boxes, by post or online except for the direct issue of a full Hong Kong driving licence.

 

Applications may be submitted via the drop-in boxes at the offices concerned between 9am and 5pm from Monday to Friday except public holidays.

 

Those who have made online appointments for renewal of a full driving licence, vehicle licence, and transfer of vehicle ownership or international driving permit can visit the Licensing Office concerned at the scheduled time.

 

To maintain social distancing to reduce the risk of spreading COVID-19, the offices concerned will continue to suspend walk-in counter services.

 

Meanwhile, the Highways Department's public services will return to normal from May 4.

 

The department will implement social distancing and precautionary measures, including the introduction of flexible working and lunch hours, meeting adjustments and stepping up the cleansing of its offices.

 

Members of the public may call the 24-hour hotline 2926 4111 or 1823 for making suggestions, enquiries or complaints. The department can also be contacted by email or fax.

Police involved in offences arrested

Police today said it is highly concerned about recent criminal offences involving police officers and that a number of these officers have been arrested.

 

Among them, officers in connection with serious offences have been or will be interdicted.

 

The force said it is furious and disappointed about the officers who are suspected of having breached the law and that the incidents have impaired public confidence in Police.

 

Police attach the utmost importance to the discipline and integrity of officers.

 

All officers, irrespective of their ranks, must abide by the law at all times.

 

Police management has zero tolerance towards any acts that breach the law or discipline by police officers, it added.

 

The force also said it attaches great importance to the integrity of its officers.

 

The Complaints & Internal Investigations Branch formulated the Integrated Integrity Management Framework to promote integrity and honesty among officers as well as to regulate their discipline and conduct.

 

To avoid similar incidents, Police said its management is reviewing the force’s internal management strategy.

 

Police reiterated that no bad element in the force can be tolerated.

 

If an officer is suspected of having breached the law or committed a breach of discipline, Police will conduct an investigation according to the established mechanism and take appropriate actions.

COVID-19 patient tests positive again

A recovered COVID-19 patient has tested positive for the virus again, the Hospital Authority announced today.

 

The authority’s Chief Manager (Quality & Standards) Dr Lau Ka-hin told a media briefing this afternoon that the patient was first admitted to Queen Mary Hospital on March 24 after having fever for a week. He subsequently tested positive for COVID-19.

 

He was discharged on April 16 after two consecutive negative tests for the virus.

 

Dr Lau said: "The patient presented to the Accident & Emergency Department of Queen Mary Hospital on May 5 because of some abdominal pain and diarrhoea.

 

"He was admitted to our hospital and was found to have a positive result for COVID-19 in the throat saliva, but the cycle threshold value is very high - nearly 36.

 

"The experts consider that this is the residual virus left in the patient’s body, which is not infective, and it is not likely to be a reinfection at this moment."

 

He added that the patient is in a stable condition.

Repression of sphingosine kinase (SK)-interacting protein (SKIP) in acute myeloid leukemia diminishes SK activity and its re-expression restores SK function [Molecular Bases of Disease]

Previous studies have shown that sphingosine kinase interacting protein (SKIP) inhibits sphingosine kinase (SK) function in fibroblasts. SK phosphorylates sphingosine producing the potent signaling molecule sphingosine-1-phosphate (S1P). SKIP gene (SPHKAP) expression is silenced by hypermethylation of its promoter in acute myeloid leukemia (AML). However, why SKIP activity is silenced in primary AML cells is unclear. Here, we investigated the consequences of SKIP down-regulation in AML primary cells and the effects of SKIP re-expression in leukemic cell lines. Using targeted ultra-HPLC-tandem MS (UPLC-MS/MS), we measured sphingolipids (including S1P and ceramides) in AML and control cells. Primary AML cells had significantly lower SK activity and intracellular S1P concentrations than control cells, and SKIP-transfected leukemia cell lines exhibited increased SK activity. These findings show that SKIP re-expression enhances SK activity in leukemia cells. Furthermore, other bioactive sphingolipids such as ceramide were also down-regulated in primary AML cells. Of note, SKIP re-expression in leukemia cells increased ceramide levels 2-fold, inactivated the key signaling protein extracellular signal-regulated kinase, and increased apoptosis following serum deprivation or chemotherapy. These results indicate that SKIP down-regulation in AML reduces SK activity and ceramide levels, an effect that ultimately inhibits apoptosis in leukemia cells. The findings of our study contrast with previous results indicating that SKIP inhibits SK function in fibroblasts and therefore challenge the notion that SKIP always inhibits SK activity.

Glucocerebrosidases catalyze a transgalactosylation reaction that yields a newly-identified brain sterol metabolite, galactosylated cholesterol [Glycobiology and Extracellular Matrices]

β-Glucocerebrosidase (GBA) hydrolyzes glucosylceramide (GlcCer) to generate ceramide. Previously, we demonstrated that lysosomal GBA1 and nonlysosomal GBA2 possess not only GlcCer hydrolase activity, but also transglucosylation activity to transfer the glucose residue from GlcCer to cholesterol to form β-cholesterylglucoside (β-GlcChol) in vitro. β-GlcChol is a member of sterylglycosides present in diverse species. How GBA1 and GBA2 mediate β-GlcChol metabolism in the brain is unknown. Here, we purified and characterized sterylglycosides from rodent and fish brains. Although glucose is thought to be the sole carbohydrate component of sterylglycosides in vertebrates, structural analysis of rat brain sterylglycosides revealed the presence of galactosylated cholesterol (β-GalChol), in addition to β-GlcChol. Analyses of brain tissues from GBA2-deficient mice and GBA1- and/or GBA2-deficient Japanese rice fish (Oryzias latipes) revealed that GBA1 and GBA2 are responsible for β-GlcChol degradation and formation, respectively, and that both GBA1 and GBA2 are responsible for β-GalChol formation. Liquid chromatography–tandem MS revealed that β-GlcChol and β-GalChol are present throughout development from embryo to adult in the mouse brain. We found that β-GalChol expression depends on galactosylceramide (GalCer), and developmental onset of β-GalChol biosynthesis appeared to be during myelination. We also found that β-GlcChol and β-GalChol are secreted from neurons and glial cells in association with exosomes. In vitro enzyme assays confirmed that GBA1 and GBA2 have transgalactosylation activity to transfer the galactose residue from GalCer to cholesterol to form β-GalChol. This is the first report of the existence of β-GalChol in vertebrates and how β-GlcChol and β-GalChol are formed in the brain.

ABC transporters control ATP release through cholesterol-dependent volume-regulated anion channel activity [Signal Transduction]

Purinergic signaling by extracellular ATP regulates a variety of cellular events and is implicated in both normal physiology and pathophysiology. Several molecules have been associated with the release of ATP and other small molecules, but their precise contributions have been difficult to assess because of their complexity and heterogeneity. Here, we report on the results of a gain-of-function screen for modulators of hypotonicity-induced ATP release using HEK-293 cells and murine cerebellar granule neurons, along with bioluminescence, calcium FLIPR, and short hairpin RNA–based gene-silencing assays. This screen utilized the most extensive genome-wide ORF collection to date, covering 90% of human, nonredundant, protein-encoding genes. We identified two ABCG1 (ABC subfamily G member 1) variants, which regulate cellular cholesterol, as modulators of hypotonicity-induced ATP release. We found that cholesterol levels control volume-regulated anion channel–dependent ATP release. These findings reveal novel mechanisms for the regulation of ATP release and volume-regulated anion channel activity and provide critical links among cellular status, cholesterol, and purinergic signaling.

Structure of an ancestral mammalian family 1B1 cytochrome P450 with increased thermostability [Enzymology]

Mammalian cytochrome P450 enzymes often metabolize many pharmaceuticals and other xenobiotics, a feature that is valuable in a biotechnology setting. However, extant P450 enzymes are typically relatively unstable, with T50 values of ∼30–40 °C. Reconstructed ancestral cytochrome P450 enzymes tend to have variable substrate selectivity compared with related extant forms, but they also have higher thermostability and therefore may be excellent tools for commercial biosynthesis of important intermediates, final drug molecules, or drug metabolites. The mammalian ancestor of the cytochrome P450 1B subfamily was herein characterized structurally and functionally, revealing differences from the extant human CYP1B1 in ligand binding, metabolism, and potential molecular contributors to its thermostability. Whereas extant human CYP1B1 has one molecule of α-naphthoflavone in a closed active site, we observed that subtle amino acid substitutions outside the active site in the ancestor CYP1B enzyme yielded an open active site with four ligand copies. A structure of the ancestor with 17β-estradiol revealed only one molecule in the active site, which still had the same open conformation. Detailed comparisons between the extant and ancestor forms revealed increases in electrostatic and aromatic interactions between distinct secondary structure elements in the ancestral forms that may contribute to their thermostability. To the best of our knowledge, this represents the first structural evaluation of a reconstructed ancestral cytochrome P450, revealing key features that appear to contribute to its thermostability.

Long noncoding RNA pncRNA-D reduces cyclin D1 gene expression and arrests cell cycle through RNA m6A modification [RNA]

pncRNA-D is an irradiation-induced 602-nt long noncoding RNA transcribed from the promoter region of the cyclin D1 (CCND1) gene. CCND1 expression is predicted to be inhibited through an interplay between pncRNA-D and RNA-binding protein TLS/FUS. Because the pncRNA-D–TLS interaction is essential for pncRNA-D–stimulated CCND1 inhibition, here we studied the possible role of RNA modification in this interaction in HeLa cells. We found that osmotic stress induces pncRNA-D by recruiting RNA polymerase II to its promoter. pncRNA-D was highly m6A-methylated in control cells, but osmotic stress reduced the methylation and also arginine methylation of TLS in the nucleus. Knockdown of the m6A modification enzyme methyltransferase-like 3 (METTL3) prolonged the half-life of pncRNA-D, and among the known m6A recognition proteins, YTH domain-containing 1 (YTHDC1) was responsible for binding m6A of pncRNA-D. Knockdown of METTL3 or YTHDC1 also enhanced the interaction of pncRNA-D with TLS, and results from RNA pulldown assays implicated YTHDC1 in the inhibitory effect on the TLS–pncRNA-D interaction. CRISPR/Cas9-mediated deletion of candidate m6A site decreased the m6A level in pncRNA-D and altered its interaction with the RNA-binding proteins. Of note, a reduction in the m6A modification arrested the cell cycle at the G0/G1 phase, and pncRNA-D knockdown partially reversed this arrest. Moreover, pncRNA-D induction in HeLa cells significantly suppressed cell growth. Collectively, these findings suggest that m6A modification of the long noncoding RNA pncRNA-D plays a role in the regulation of CCND1 gene expression and cell cycle progression.

AIG1 and ADTRP are endogenous hydrolases of fatty acid esters of hydroxy fatty acids (FAHFAs) in mice [Metabolism]

Fatty acid esters of hydroxy fatty acids (FAHFAs) are a newly discovered class of signaling lipids with anti-inflammatory and anti-diabetic properties. However, the endogenous regulation of FAHFAs remains a pressing but unanswered question. Here, using MS-based FAHFA hydrolysis assays, LC-MS–based lipidomics analyses, and activity-based protein profiling, we found that androgen-induced gene 1 (AIG1) and androgen-dependent TFPI-regulating protein (ADTRP), two threonine hydrolases, control FAHFA levels in vivo in both genetic and pharmacologic mouse models. Tissues from mice lacking ADTRP (Adtrp-KO), or both AIG1 and ADTRP (DKO) had higher concentrations of FAHFAs particularly isomers with the ester bond at the 9th carbon due to decreased FAHFA hydrolysis activity. The levels of other lipid classes were unaltered indicating that AIG1 and ADTRP specifically hydrolyze FAHFAs. Complementing these genetic studies, we also identified a dual AIG1/ADTRP inhibitor, ABD-110207, which is active in vivo. Acute treatment of WT mice with ABD-110207 resulted in elevated FAHFA levels, further supporting the notion that AIG1 and ADTRP activity control endogenous FAHFA levels. However, loss of AIG1/ADTRP did not mimic the changes associated with pharmacologically administered FAHFAs on extent of upregulation of FAHFA levels, glucose tolerance, or insulin sensitivity in mice, indicating that therapeutic strategies should weigh more on FAHFA administration. Together, these findings identify AIG1 and ADTRP as the first endogenous FAHFA hydrolases identified and provide critical genetic and chemical tools for further characterization of these enzymes and endogenous FAHFAs to unravel their physiological functions and roles in health and disease.

LSU Health study suggests nicotine exposure alone leads to pulmonary hypertension

(Louisiana State University Health Sciences Center) A study conducted at LSU Health New Orleans has shown for the first time that chronic exposure to inhaled nicotine alone increases blood pressure in both the body's general circulation and in the lungs that can lead to pulmonary hypertension. The study also found that nicotine-induced pulmonary hypertension is accompanied by changes in the size, shape and function (remodeling) of the blood vessels in the lung and the right lower chamber of the heart.

The Agriculture, Fisheries & Conservation Department today said genetic sequencing tests are being carried out on the pet dog of a confirmed COVID-19 patient which could reveal if the virus has mutated or not.

 

The department's Assistant Director (Inspection & Quarantine) Dr Thomas Sit told a press briefing this afternoon that the coronavirus is what is known as an RNA virus which eventually mutates.

 

“That is why the University of Hong Kong School of Public Health needs to do a genetic analysis to compare this dog’s genetic sequencing to the virus isolated from the patient so that they can compare. So if it is totally identical, then there is no mutation. The testing is still ongoing.”

 

Dr Sit reiterated that international experts agreed that the dog has a low level of infection, despite its blood tests not being ready yet.

 

“From the first sample to our last sample tested, it has already (been) six days. The dog’s nasal or oral mechanism, their secretion they should have - if contaminated - they should have a way to clean the virus, it would not stay for that long if it was just a contamination.

 

“I think it will take at least five or seven days for the blood results because it is not an easy test, it is not a quick test. We need to grow a virus and then neutralise the serum, so it takes some time.”

 

He added that it was too early to say whether animal-to-animal transmission was a possibility.

 

“At this stage, we do not have enough data to have a 100% answer as to whether it is infectious to other dogs or not. But if the dog’s owner is positive, it is better to take precautionary measures to prevent onward transmission.”

 

Dr Sit also advised dog owners to wash their hands, wear gloves and try to stop their dogs from licking their surroundings to prevent the virus from spreading further.

Dog tests positive for COVID-19

The Agriculture, Fisheries & Conservation Department (AFCD) today said a pet dog has tested positive for the COVID-19 virus. 

The case involves a German Shepherd dog whose owners live in Pok Fu Lam. This follows an earlier case in which a 17-year-old Pomeranian dog tested weak positive during repeated tests for the virus.

  

When the German Shepherd's owner was confirmed with COVID-19, it was sent for quarantine with another mixed breed dog from the same residence to the animal keeping facility at the Hong Kong-Zhuhai-Macao Bridge's Hong Kong Port yesterday.

 

No positive results were obtained from the mixed breed dog and neither dog has shown any signs of the disease.

 

The department will continue to closely monitor both dogs and conduct repeated tests on them.

 

It strongly advises that mammalian pet animals including dogs and cats from households with people confirmed as infected with COVID-19, or close contacts of COVID-19 infected people, should be put under quarantine in AFCD facilities.

 

The department emphasised that there is currently no evidence pet animals can be a source of COVID-19 for humans or that this virus can cause the disease in dogs.

 

Pet owners are reminded to maintain good hygiene practices and under no circumstances should they abandon their pets.

ESO instrument finds closest black hole to Earth

(ESO) Astronomers have discovered a black hole lying just 1,000 light-years from Earth. The black hole is closer to our solar system than any other found to date and forms part of a triple system that can be seen with the naked eye. The astronomers found evidence for the invisible object by tracking its two companion stars using the MPG/ESO 2.2-meter telescope at ESO's La Silla Observatory in Chile. They say this system could just be the tip of the iceberg.
