Nestin Selectively Facilitates the Phosphorylation of the Lissencephaly-Linked Protein Doublecortin (DCX) by cdk5/p35 to Regulate Growth Cone Morphology and Sema3a Sensitivity in Developing Neurons

Nestin, an intermediate filament protein widely used as a marker of neural progenitors, was recently found to be expressed transiently in developing cortical neurons in culture and in developing mouse cortex. In young cortical cultures, nestin regulates axonal growth cone morphology. In addition, nestin, which is known to bind the neuronal cdk5/p35 kinase, affects responses to axon guidance cues upstream of cdk5, specifically, to Sema3a. Changes in growth cone morphology require rearrangements of cytoskeletal networks, and changes in microtubules and actin filaments are well studied. In contrast, the roles of intermediate filament proteins in this process are poorly understood, even in cultured neurons. Here, we investigate the molecular mechanism by which nestin affects growth cone morphology and Sema3a sensitivity. We find that nestin selectively facilitates the phosphorylation of the lissencephaly-linked protein doublecortin (DCX) by cdk5/p35, but the phosphorylation of other cdk5 substrates is not affected by nestin. We uncover that this substrate selectivity is based on the ability of nestin to interact with DCX, but not with other cdk5 substrates. Nestin thus creates a selective scaffold for DCX with activated cdk5/p35. Last, we use cortical cultures derived from Dcx KO mice to show that the effects of nestin on growth cone morphology and on Sema3a sensitivity are DCX-dependent, thus suggesting a functional role for the DCX-nestin complex in neurons. We propose that nestin changes growth cone behavior by regulating the intracellular kinase signaling environment in developing neurons. The sex of animal subjects is unknown.

SIGNIFICANCE STATEMENT Nestin, an intermediate filament protein highly expressed in neural progenitors, was recently identified in developing neurons where it regulates growth cone morphology and responsiveness to the guidance cue Sema3a. Church in South Korea growing, slowly

CNA Staff, Apr 27, 2020 / 11:30 am (CNA).- The number of Catholics in South Korea increased by less than 50,000 in 2019, continuing a trend of slowing growth after a peak following the 2014 apostolic visit by Pope Francis to the country. 

According to the Catholic Bishops' Conference of Korea, there are 5.91 million Catholics in South Korea’s 16 dioceses; an increase of 48,000 over to 2018’s total. Catholics make up 11% of the national population. 

Overall, the number of Catholics in the country increased by 0.8%, which is slightly lower than last year’s increase of 0.9%. In recent years, the Catholic population in South Korea has grown by an average of 1% each year. 

Like many countries, South Korea’s Catholic population is aging. About one in five South Korean Catholics are over the age of 65, and only 8.5% of Catholics are age 19 or under. A total of 14% of priests are over the age of 65.

South Korea’s flock saw the largest overall percentage increase in 2014, when it increased by 2.2%. Pope Francis visited the country in August of that year, the third visit by a pope to the country. Pope John Paul II visited South Korea in 1984 and again in 1989, when the country hosted the 44th International Eucharistic Congress.

These numbers come as South Korea is grappling with an outbreak of COVID-19, a disease which has seen much of its spread in the nation come from a single member of the Shincheonji Church of Jesus. About half of South Korea’s nearly 11,000 confirmed coronavirus infections stem from “Patient 31,” a member of that church who did not abide by isolation rules after coming down with COVID-19. 

The Catholic Bishops’ Association of Korea refers to Shincheonji Church of Jesus as a “pseudo-religious organization” and a “cult.” The church was founded in 1980 by a man who believes that he is the second coming of Jesus. 

In 2017, the conference created the "Korean Catholic Task Force on Newly-risen Religions” specifically to combat the growing popularity of the Shincheonji Church of Jesus. 

Another source of outbreak were 30 Catholic pilgrims who tested positive for the illness after returning to South Korea from a trip to the Holy Land. Korean Air Lines restricted travel to Israel after these infections were discovered.  

South Korea suspended the public celebration of Mass in late February, and re-opened churches--albeit with strict social distancing requirements--on April 26.

Impaired fetal growth is an independent risk factor for cardiovascular diseases in adulthood and is associated with arterial wall thickening, a noninvasive measure of subclinical atherosclerosis, in early childhood. No preventive strategy has been identified.

Dietary omega-3 fatty acid supplementation in early childhood prevented the association of impaired fetal growth with arterial wall thickening, suggesting that this early-life intervention may mitigate the risk of cardiovascular disease in those with impaired fetal growth. Early or Delayed Enteral Feeding for Preterm Growth-Restricted Infants: A Randomized Trial

Preterm, growth-restricted infants are at high risk of necrotizing enterocolitis (NEC). NEC occurs most frequently in infants who have received enteral feeds. It is common practice to delay introduction of enteral feeds in these infants.

Early introduction of enteral feeds results in earlier achievement of full enteral feeding. Early feeding is not associated with a higher risk of NEC. Delayed feeding is associated with a higher risk of cholestasis. (Read the full article)

Infantile hemangiomas have a period of rapid growth in early infancy. Most hemangioma growth is completed by 5 months of age, but the majority of patients are not seen by a specialist until after the growth phase is complete.

Growth in Children With Congenital Heart Disease

Children with congenital heart disease (CHD) are at increased risk for poor growth. Several factors may play a role in poor growth, including feeding difficulties, increased caloric requirements, and the effects of cardiac lesions on growth regulation.

Growth of Extremely Preterm Survivors From Birth to 18 Years of Age Compared With Term Controls

Children born at very low birth weights have significant catch-up weight gain but differences in height remain. Their BMI, however, tends not to be higher than expected. Data are lacking regarding representative cohorts, defined by gestation and compared with contemporaneous controls.

In a geographic cohort of extremely preterm participants followed until age 18, compared with term controls, weight differences diminish over time, and height differences persist. Growth Outcomes of Weight Faltering in Infancy in ALSPAC

Studies of clinically derived samples of infants with failure to thrive have reported that children remain shorter and lighter than their peers at school-age. Enhanced weight gain ("catch-up") in small infants has been linked to subsequent obesity.

Infants with early weight faltering caught up in weight by 2 years, but height gain remained disproportionally slow. Those with weight faltering later in infancy remained shorter and lighter throughout childhood. Anthropometric outcomes of both groups were within population norms at 13 years. Cerebral Palsy and Growth Failure at 6 to 7 Years

Surviving infants with hypoxic ischemic encephalopathy (HIE) treated with hypothermia have decreased rates of CP in childhood. CP is associated with increased risk of slow growth.

Postnatal Head Growth in Preterm Infants: A Randomized Controlled Parenteral Nutrition Study

Preterm infants dependent on parenteral nutrition are vulnerable to deficits in early postnatal nutritional intake. This coincides with a period of suboptimal head growth. Observational studies indicate that poor nutritional intake is associated with suboptimal head growth and neurodevelopmental outcome.

This study provides randomized controlled trial evidence that head growth failure in the first 4 weeks of life can be ameliorated with early nutritional intervention. Symmetrical and Asymmetrical Growth Restriction in Preterm-Born Children

Fetal growth restriction, particularly in preterm children, is associated with delayed development and poor growth. Knowledge about the consequences of fetal growth restriction if classified by symmetry is lacking, especially in preterm children.

Growth Patterns of Large for Gestational Age Children up to Age 4 Years

Preterm (PT) birth is negatively associated with growth. Particularly small for gestational age PT infants are at risk for delays in growth, whereas knowledge about the consequences regarding growth of large for gestational age PT birth is lacking.

During infancy, growth in height, weight, and head circumference of large for gestational age PT infants was well balanced and sufficient. Growth After Adenotonsillectomy for Obstructive Sleep Apnea: An RCT

Growth failure has been frequently reported in children who have obstructive sleep apnea syndrome (OSAS) owing to adenotonsillar hypertrophy. Adenotonsillectomy (AT) has been reported to accelerate weight gain in children who have OSAS in nonrandomized uncontrolled studies.

This randomized controlled trial of AT for pediatric OSAS demonstrated significantly greater weight increases 7 months after AT in all weight categories. AT normalizes weight in children who have failure to thrive, but increases risk for obesity in overweight children. (Read the full article)

Brain Injury and Altered Brain Growth in Preterm Infants: Predictors and Prognosis

Term MRI can assist in identifying the nature and extent of brain injury in preterm infants. However, brain injury detected by MRI does not fully account for neurodevelopmental impairments, particularly cognitive and behavioral impairments, common in preterm survivors.

In addition to brain injury, an assessment of brain growth by using one-dimensional measurements on MRI is helpful for predicting neurodevelopment.
