Of the approved plans, nine were for apartment and apartment-commercial developments, one was for commercial development, one was for factory and industrial developments, and two were for community services developments. Consent was given for works to start on two building projects, which will provide 77,942 sq m of gross floor area for non-domestic use. The department also issued 14 occupation permits - four on Hong Kong Island, one in Kowloon and nine in the New Territories. Buildings certified for occupation have 149,306 sq m of gross floor area for domestic use involving 2,912 units, and 67,594 sq m for non-domestic use. Mr Hui visited the New Companies Section, the Public Search Section and the Document Management Section at the registry and spoke with staff there to learn about their work conditions and the services that they provide. He said: “The COVID-19 pandemic has dealt a heavy blow to Hong Kong's overall economy. “To help enterprises cope with their operating pressure amid the economic downturn, the Financial Secretary announced in the 2020-21 Budget the waiver of registration fees for annual returns, except for late delivery, charged by the CR for two years. “And with a view to encouraging the wider use of the CR's electronic services, we also propose to reduce the fees payable in relation to the incorporation of companies, including registration of non-Hong Kong companies, through electronic means by 10%." The Companies (Fees) (Amendment) Regulation 2020 gazetted today will be tabled at the Legislative Council for negative vetting on May 13 for the waiver and reduction to take effect from October 1. The waiver of registration fees for annual returns will benefit about 1.4 million companies. Mr Hui added that he was pleased that the CR has been providing electronic services for filing of documents and company searches. He appealed to the department to adopt wider use of technology, adding that a business-friendly environment is needed more than ever in the process of economic recovery. Given the stabilising epidemic situation, the Government has conducted a health risk assessment and will allow premises, including fitness centres, to resume operations, Mr Tsui said, adding that he hopes the fitness industry will soon regain vitality. The Home Affairs Bureau commissioned the association to assist in implementing the scheme, which opened for applications on May 4. As of May 7, the association received 397 applications, of which more than half of them have been initially found to be eligible, involving subsidies of about $20 million. ui Proline-rich 11 (PRR11) drives F-actin assembly by recruiting the actin-related protein 2/3 complex in human non-small cell lung carcinoma [DNA and Chromosomes] By www.jbc.org Published On :: 2020-04-17T00:06:05-07:00 The actin cytoskeleton is extremely dynamic and supports diverse cellular functions in many physiological and pathological processes, including tumorigenesis. However, the mechanisms that regulate the actin-related protein 2/3 (ARP2/3) complex and thereby promote actin polymerization and organization in cancer cells are not well-understood. We previously implicated the proline-rich 11 (PRR11) protein in lung cancer development. In this study, using immunofluorescence staining, actin polymerization assays, and siRNA-mediated gene silencing, we uncovered that cytoplasmic PRR11 is involved in F-actin polymerization and organization. We found that dysregulation of PRR11 expression results in F-actin rearrangement and nuclear instability in non-small cell lung cancer cells. Results from molecular mechanistic experiments indicated that PRR11 associates with and recruits the ARP2/3 complex, facilitates F-actin polymerization, and thereby disrupts the F-actin cytoskeleton, leading to abnormal nuclear lamina assembly and chromatin reorganization. Inhibition of the ARP2/3 complex activity abolished irregular F-actin polymerization, lamina assembly, and chromatin reorganization due to PRR11 overexpression. Notably, experiments with truncated PRR11 variants revealed that PRR11 regulates F-actin through different regions. We found that deletion of either the N or C terminus of PRR11 abrogates its effects on F-actin polymerization and nuclear instability and that deletion of amino acid residues 100–184 or 100–200 strongly induces an F-actin structure called the actin comet tail, not observed with WT PRR11. Our findings indicate that cytoplasmic PRR11 plays an essential role in regulating F-actin assembly and nuclear stability by recruiting the ARP2/3 complex in human non-small cell lung carcinoma cells. Current findings support the idea that for EsxA to interact with host membranes, EsxA must dissociate from EsxB at low pH. However, the molecular mechanism by which the EsxA:B heterodimer separates is not clear. In the present study, using liposome-leakage and cytotoxicity assays, LC-MS/MS–based proteomics, and CCF-4 FRET analysis, we obtained evidence that the Nα-acetylation of the Thr-2 residue on EsxA, a post-translational modification that is present in mycobacteria but absent in Escherichia coli, is required for the EsxA:B separation. Substitutions at Thr-2 that precluded Nα-acetylation inhibited the heterodimer separation and hence prevented EsxA from interacting with the host membrane, resulting in attenuated mycobacterial cytosolic translocation and virulence. Molecular dynamics simulations revealed that at low pH, the Nα-acetylated Thr-2 makes direct and frequent “bind-and-release” contacts with EsxB, which generates a force that pulls EsxB away from EsxA. In summary, our findings provide evidence that the Nα-acetylation at Thr-2 of EsxA facilitates dissociation of the EsxA:B heterodimer required for EsxA membrane permeabilization and mycobacterial cytosolic translocation and virulence. ui MtrP, a putative methyltransferase in Corynebacteria, is required for optimal membrane transport of trehalose mycolates [Lipids] By www.jbc.org Published On :: 2020-05-01T00:06:09-07:00 Pathogenic bacteria of the genera Mycobacterium and Corynebacterium cause severe human diseases such as tuberculosis (Mycobacterium tuberculosis) and diphtheria (Corynebacterium diphtheriae). The cells of these species are surrounded by protective cell walls rich in long-chain mycolic acids. These fatty acids are conjugated to the disaccharide trehalose on the cytoplasmic side of the bacterial cell membrane. They are then transported across the membrane to the periplasm where they act as donors for other reactions. We have previously shown that transient acetylation of the glycolipid trehalose monohydroxycorynomycolate (hTMCM) enables its efficient transport to the periplasm in Corynebacterium glutamicum and that acetylation is mediated by the membrane protein TmaT. Here, we show that a putative methyltransferase, encoded at the same genetic locus as TmaT, is also required for optimal hTMCM transport. Deletion of the C. glutamicum gene NCgl2764 (Rv0224c in M. tuberculosis) abolished acetyltrehalose monocorynomycolate (AcTMCM) synthesis, leading to accumulation of hTMCM in the inner membrane and delaying its conversion to trehalose dihydroxycorynomycolate (h2TDCM). Complementation with NCgl2764 normalized turnover of hTMCM to h2TDCM. In contrast, complementation with NCgl2764 derivatives mutated at residues essential for methyltransferase activity failed to rectify the defect, suggesting that NCgl2764/Rv0224c encodes a methyltransferase, designated here as MtrP. Comprehensive analyses of the individual mtrP and tmaT mutants and of a double mutant revealed strikingly similar changes across several lipid classes compared with WT bacteria. These findings indicate that both MtrP and TmaT have nonredundant roles in regulating AcTMCM synthesis, revealing additional complexity in the regulation of trehalose mycolate transport in the Corynebacterineae. Recent reports indicate that, in the presence of crowding agents, tau can undergo liquid–liquid phase separation (LLPS), forming highly dynamic liquid droplets. Here, using recombinantly expressed proteins, turbidimetry, fluorescence microscopy imaging, and fluorescence recovery after photobleaching (FRAP) assays, we show that the divalent transition metal zinc strongly promotes this process, shifting the equilibrium phase boundary to lower protein or crowding agent concentrations. We observed no tau LLPS-promoting effect for any other divalent transition metal ions tested, including Mn2+, Fe2+, Co2+, Ni2+, and Cu2+. We also demonstrate that multiple zinc-binding sites on tau are involved in the LLPS-promoting effect and provide insights into the mechanism of this process. Zinc concentration is highly elevated in AD brains, and this metal ion is believed to be an important player in the pathogenesis of this disease. Thus, the present findings bring a new dimension to understanding the relationship between zinc homeostasis and the pathogenic process in AD and related neurodegenerative disorders. Full Article

Walls, both inside and out, were worn down, staircases were broken, and with electrical installation panels exposed on the outside. But with the help of various subsidy schemes launched by the Government and Urban Renewal Authority, older buildings are being given a new lease of life. The schemes include Operation Building Bright 2.0 (OBB 2.0), the Fire Safety Improvement Works Subsidy Scheme (FSWS) and the Lift Modernisation Subsidy Scheme (LIMSS). Helping handOctogenarian owner-occupiers Leung Ting-lam and Chu Lai-chun have lived in this building on Fa Yuen Street for more than two decades. They welcome the improvements, which include a new metal gate and lift. “Modernising the lift was done so quickly. It only took a few months and that was the most satisfying part,” said Mr Leung. Ms Chu agreed that the lift no longer breaks down so regularly. It cost $800,000 just to modernise the lift. Add to that the other repair and maintenance works, and the total cost exceeded $3 million. Those who own a flat had to shoulder tens of thousands of dollars in the 10-storey building containing 19 units. For seniors on a fixed income, it is not easy to cover the full cost of repair works. With the government subsidies, they no longer have to worry about the financial burden of upgrading their buildings on their own. Popular servicesThe lift modernisation work under the LIMSS is assisted by the Electrical & Mechanical Services Department. The department’s Assistant Director Raymond Poon said the subsidy can cover up to 60% of the total cost of the works - plus a consultation fee - with a cap of $500,000 per lift for eligible buildings. For elderly owner-occupiers, they can receive the full cost of relevant works capped at $50,000 per domestic unit. Up until August 1, the department received around 1,171 applications for the scheme involving nearly 5,000 lifts in the first round of applications. This far exceeded the quota of 1,400 lifts set for the round, a response Mr Poon described as overwhelming. Apart from the LIMSS, owner-occupiers can also apply for OBB 2.0 and other schemes to cover maintenance costs. In one 58-year-old Hung Hom building, residents had to spend more than $800,000 to paint the common areas, replace above-ground drainage pipework and other structural upgrades. The building’s owners’ corporation has applied for several funding schemes to get some financial relief, including OBB 2.0. Under this scheme, elderly recipients can receive the full cost of the work, subject to a cap of $50,000. Other owner-occupiers can receive 80%, capped at $40,000. As of August, the owners or owners’ corporations of 479 Category One buildings were prepared to carry out the prescribed inspection and repair works for the common areas of their buildings under OBB 2.0. They can do so on a voluntary basis to comply with the Mandatory Building Inspection Scheme (MBIS) statutory notices. These buildings are in 13 districts. Among them, Yau Tsim Mong has the largest number of applications, followed by Kowloon City and Sham Shui Po. More help aheadIn the future, another $3 billion will be injected into OBB 2.0. This means a total of $6 billion will be used to benefit 5,000 buildings. Development Bureau Principal Assistant Secretary Jasmine Choi said they have received feedback from the community which hopes the operation will eventually allow younger buildings to join the rehabilitation schemes. Upon review, buildings between 40 and 49 years old with an outstanding MBIS statutory notice not yet complied with will be accepted in the next round of applications. ui MtrP, a putative methyltransferase in Corynebacteria, is required for optimal membrane transport of trehalose mycolates [Lipids] By feedproxy.google.com Published On :: 2020-05-01T00:06:09-07:00 Pathogenic bacteria of the genera Mycobacterium and Corynebacterium cause severe human diseases such as tuberculosis (Mycobacterium tuberculosis) and diphtheria (Corynebacterium diphtheriae). The cells of these species are surrounded by protective cell walls rich in long-chain mycolic acids. These fatty acids are conjugated to the disaccharide trehalose on the cytoplasmic side of the bacterial cell membrane. They are then transported across the membrane to the periplasm where they act as donors for other reactions. We have previously shown that transient acetylation of the glycolipid trehalose monohydroxycorynomycolate (hTMCM) enables its efficient transport to the periplasm in Corynebacterium glutamicum and that acetylation is mediated by the membrane protein TmaT. Here, we show that a putative methyltransferase, encoded at the same genetic locus as TmaT, is also required for optimal hTMCM transport. Deletion of the C. glutamicum gene NCgl2764 (Rv0224c in M. tuberculosis) abolished acetyltrehalose monocorynomycolate (AcTMCM) synthesis, leading to accumulation of hTMCM in the inner membrane and delaying its conversion to trehalose dihydroxycorynomycolate (h2TDCM). Complementation with NCgl2764 normalized turnover of hTMCM to h2TDCM. In contrast, complementation with NCgl2764 derivatives mutated at residues essential for methyltransferase activity failed to rectify the defect, suggesting that NCgl2764/Rv0224c encodes a methyltransferase, designated here as MtrP. Comprehensive analyses of the individual mtrP and tmaT mutants and of a double mutant revealed strikingly similar changes across several lipid classes compared with WT bacteria. These findings indicate that both MtrP and TmaT have nonredundant roles in regulating AcTMCM synthesis, revealing additional complexity in the regulation of trehalose mycolate transport in the Corynebacterineae.
