Simian Immunodeficiency Virus-Infected Memory CD4+ T Cells Infiltrate to the Site of Infected Macrophages in the Neuroparenchyma of a Chronic Macaque Model of Neurological Complications of AIDS

ABSTRACT

Simian immunodeficiency virus (SIV)-infected nonhuman primates can serve as a relevant model for AIDS neuropathogenesis. Current SIV-induced encephalitis (SIVE)/neurological complications of AIDS (neuroAIDS) models are generally associated with rapid progression to neuroAIDS, which does not reflect the tempo of neuroAIDS progression in humans. Recently, we isolated a neuropathogenic clone, SIVsm804E-CL757 (CL757), obtained from an SIV-infected rhesus macaque (RM). CL757 causes a more protracted progression to disease, inducing SIVE in 50% of inoculated animals, with high cerebral spinal fluid viral loads, multinucleated giant cells (MNGCs), and perivascular lymphocytic cuffing in the central nervous system (CNS). This latter finding is reminiscent of human immunodeficiency virus (HIV) encephalitis in humans but not generally observed in rapid progressor animals with neuroAIDS. Here, we studied which subsets of cells within the CNS were targeted by CL757 in animals with neurological symptoms of SIVE. Immunohistochemistry of brain sections demonstrated infiltration of CD4+ T cells (CD4) and macrophages (Ms) to the site of MNGCs. Moreover, an increase in mononuclear cells isolated from the brain tissues of RMs with SIVE correlated with increased cerebrospinal fluid (CSF) viral load. Subset analysis showed a specific increase in brain CD4+ memory T cells (Br-mCD4), brain-Ms (Br-Ms), and brain B cells (Br-B cells). Both Br-mCD4s and Br-Ms harbored replication-competent viral DNA, as demonstrated by virus isolation by coculture. However, only in animals exhibiting SIVE/neuroAIDS was virus isolated from Br-Ms. These findings support the use of CL757 to study the pathogenesis of AIDS viruses in the central nervous system and indicate a previously unanticipated role of CD4s cells as a potential reservoir in the brain.

IMPORTANCE While the use of combination antiretroviral therapy effectively suppresses systemic viral replication in the body, neurocognitive disorders as a result of HIV infection of the central nervous system (CNS) remain a clinical problem. Therefore, the use of nonhuman primate models is necessary to study mechanisms of neuropathogenesis. The neurotropic, molecular clone SIVsm804E-CL757 (CL757) results in neuroAIDS in 50% of infected rhesus macaques approximately 1 year postinfection. Using CL757-infected macaques, we investigate disease progression by examining subsets of cells within the CNS that were targeted by CL757 and could potentially serve as viral reservoirs. By isolating mononuclear cells from the brains of SIV-infected rhesus macaques with and without encephalitis, we show that immune cells invade the neuroparenchyma and increase in number in the CNS in animals with SIV-induced encephalitis (SIVE). Of these cells, both brain macrophages and brain memory CD4+ T cells harbor replication-competent SIV DNA; however, only brain CD4+ T cells harbored SIV DNA in animals without SIVE. These findings support use of CL757 as an important model to investigate disease progression in the CNS and as a model to study virus reservoirs in the CNS.

A flexible network of vimentin intermediate filaments promotes migration of amoeboid cancer cells through confined environments [Cell Biology]

Tumor cells can spread to distant sites through their ability to switch between mesenchymal and amoeboid (bleb-based) migration. Because of this difference, inhibitors of metastasis must account for each migration mode. However, the role of vimentin in amoeboid migration has not been determined. Because amoeboid leader bleb–based migration (LBBM) occurs in confined spaces and vimentin is known to strongly influence cell-mechanical properties, we hypothesized that a flexible vimentin network is required for fast amoeboid migration. To this end, here we determined the precise role of the vimentin intermediate filament system in regulating the migration of amoeboid human cancer cells. Vimentin is a classic marker of epithelial-to-mesenchymal transition and is therefore an ideal target for a metastasis inhibitor. Using a previously developed polydimethylsiloxane slab–based approach to confine cells, RNAi-based vimentin silencing, vimentin overexpression, pharmacological treatments, and measurements of cell stiffness, we found that RNAi-mediated depletion of vimentin increases LBBM by ∼50% compared with control cells and that vimentin overexpression and simvastatin-induced vimentin bundling inhibit fast amoeboid migration and proliferation. Importantly, these effects were independent of changes in actomyosin contractility. Our results indicate that a flexible vimentin intermediate filament network promotes LBBM of amoeboid cancer cells in confined environments and that vimentin bundling perturbs cell-mechanical properties and inhibits the invasive properties of cancer cells.

The identification and mitigation of geohazards using shallow airborne engineering geophysics and land-based geophysics for brown- and greenfield road investigations

South Africa is a mineral-rich country with a diverse geology and a long history of mining. The rich history of mining activities includes the extraction of coal from the Ecca Group Sediments of the Karoo Supergroup (250 Ma), gold and uranium from the Witwatersrand Supergroup (2900 Ma), as well as platinum, uranium, tin and lead from the layered Bushveld Igneous Complex (BIC) (2150 Ma). The extraction of gold, copper, tin, lead and rare earth minerals also took place in the Archean rocks of Swazium age (3500–3000 Ma). The historical mining records have either not been accurately recorded or have been lost over time. This has resulted in significant geohazard risk during infrastructure development, especially in and around historical mining towns, such as Johannesburg and Ermelo. These geohazard risks require careful appraisal and quantification prior to any infrastructure design or construction.

This case study aims to set out the development aspects of the Multi-Faceted Geophysical Modelling Systems approach, which was used by the South African National Roads Agency SOC Ltd (SANRAL) during an investigation of undermined ground for the historical coal-mining town of Ermelo in South Africa. The N11/N2 ring road was planned to go around Ermelo to ensure mobility between major routes, whilst still maintaining town access.

The systems approach used a combination of airborne geophysics (Versatile Time Domain Electromagnetic System (VTEMTM) and magnetics), generally used in mining exploration, land-based and borehole geophysics, borehole water testing, and ground-truthing. The approach was continuous and iterative, building on the data at hand and reducing unnecessary investigations while eliminating the possibility of anomalies being missed, as in the case of conventional discrete drilling. The investigation ensured that 100% of the route was comprehensively investigated with a high confidence in the geological and geophysical data, and concomitant mitigation of infrastructure risk.

The Multi-Faceted Geophysical Modelling Systems approach was successfully used to identify a previously unknown 1 x 1 m mining stope cavity at 90 m depth and a 3 x 5 m access tunnel at 24 m depth in a timely and cost-effective manner. Seven reverse-circulation percussion boreholes confirmed the structural integrity of these underground cavities, as well as the structural geology along the centreline. Based on the great success achieved in identifying shallow anomalies, this Multi-Faceted Geophysical Modelling Systems approach is now being considered for field trails on the dolomitic formations and the Wild Coast greenfields road project where there are large historical slumps and many fault lines.

18F-FAC PET Visualizes Brain-Infiltrating Leukocytes in a Mouse Model of Multiple Sclerosis

Brain-infiltrating leukocytes contribute to multiple sclerosis (MS) and autoimmune encephalomyelitis and likely play a role in traumatic brain injury, seizure, and stroke. Brain-infiltrating leukocytes are also primary targets for MS disease-modifying therapies. However, no method exists for noninvasively visualizing these cells in a living organism. 1-(2'-deoxy-2'-18F-fluoroarabinofuranosyl) cytosine (18F-FAC) is a PET radiotracer that measures deoxyribonucleoside salvage and accumulates preferentially in immune cells. We hypothesized that 18F-FAC PET could noninvasively image brain-infiltrating leukocytes. Methods: Healthy mice were imaged with 18F-FAC PET to quantify if this radiotracer crosses the blood–brain barrier (BBB). Experimental autoimmune encephalomyelitis (EAE) is a mouse disease model with brain-infiltrating leukocytes. To determine whether 18F-FAC accumulates in brain-infiltrating leukocytes, EAE mice were analyzed with 18F-FAC PET, digital autoradiography, and immunohistochemistry, and deoxyribonucleoside salvage activity in brain-infiltrating leukocytes was analyzed ex vivo. Fingolimod-treated EAE mice were imaged with 18F-FAC PET to assess if this approach can monitor the effect of an immunomodulatory drug on brain-infiltrating leukocytes. PET scans of individuals injected with 2-chloro-2'-deoxy-2'-18F-fluoro-9-β-d-arabinofuranosyl-adenine (18F-CFA), a PET radiotracer that measures deoxyribonucleoside salvage in humans, were analyzed to evaluate whether 18F-CFA crosses the human BBB. Results: 18F-FAC accumulates in the healthy mouse brain at levels similar to 18F-FAC in the blood (2.54 ± 0.2 and 3.04 ± 0.3 percentage injected dose per gram, respectively) indicating that 18F-FAC crosses the BBB. EAE mice accumulate 18F-FAC in the brain at 180% of the levels of control mice. Brain 18F-FAC accumulation localizes to periventricular regions with significant leukocyte infiltration, and deoxyribonucleoside salvage activity is present at similar levels in brain-infiltrating T and innate immune cells. These data suggest that 18F-FAC accumulates in brain-infiltrating leukocytes in this model. Fingolimod-treated EAE mice accumulate 18F-FAC in the brain at 37% lower levels than control-treated EAE mice, demonstrating that 18F-FAC PET can monitor therapeutic interventions in this mouse model. 18F-CFA accumulates in the human brain at 15% of blood levels (0.08 ± 0.01 and 0.54 ± 0.07 SUV, respectively), indicating that 18F-CFA does not cross the BBB in humans. Conclusion: 18F-FAC PET can visualize brain-infiltrating leukocytes in a mouse MS model and can monitor the response of these cells to an immunomodulatory drug. Translating this strategy into humans will require exploring additional radiotracers.

Histologically Confirmed Diagnostic Efficacy of 18F-rhPSMA-7 PET for N-Staging of Patients with Primary High-Risk Prostate Cancer

18F-rhPSMA-7 (radiohybrid prostate-specific membrane antigen [PSMA]) is a novel ligand for PET imaging. Here, we present data from a retrospective analysis using PET/CT and PET/MRI examinations to investigate the efficacy of 18F-rhPSMA-7 PET for primary N-staging of patients with prostate cancer (PC) compared with morphologic imaging (CT or MRI) and validated by histopathology. Methods: Data from 58 patients with high-risk PC (according to the D’Amico criteria) who were staged with 18F-rhPSMA-7 PET/CT or PET/MRI at our institution between July 2017 and June 2018 were reviewed. The patients had a median prescan prostate-specific antigen value of 12.2 ng/mL (range, 1.2–81.6 ng/mL). The median injected activity of 18F-rhPSMA-7 was 327 MBq (range, 132–410 MBq), with a median uptake time of 79.5 min (range, 60–153 min). All patients underwent subsequent radical prostatectomy and extended pelvic lymph node dissection. The presence of lymph node metastases was determined by an experienced reader independently for both the PET and the morphologic datasets using a template-based analysis on a 5-point scale. Patient-level and template-based results were both compared with histopathologic findings. Results: Lymph node metastases were present in 18 patients (31.0%) and were located in 52 of 375 templates (13.9%). Receiver-operating-characteristic analyses showed 18F-rhPSMA-7 PET to perform significantly better than morphologic imaging on both patient-based and template-based analyses (areas under curve, 0.858 vs. 0.649 [P = 0.012] and 0.765 vs. 0.589 [P < 0.001], respectively). On patient-based analyses, the sensitivity, specificity, and accuracy of 18F-rhPSMA-7 PET were 72.2%, 92.5%, and 86.2%, respectively, and those of morphologic imaging were 50.0%, 72.5%, and 65.5%, respectively. On template-based analyses, the sensitivity, specificity, and accuracy of 18F-rhPSMA-7 PET were 53.8%, 96.9%, and 90.9%, respectively, and those of morphologic imaging were 9.6%, 95.0%, and 83.2%, respectively. Conclusion: 18F-rhPSMA-7 PET is superior to morphologic imaging for N-staging of high-risk primary PC. The efficacy of 18F-rhPSMA-7 is similar to published data for 68Ga-PSMA-11.

Comparative Whole-Genome Phylogeny of Animal, Environmental, and Human Strains Confirms the Genogroup Organization and Diversity of the Stenotrophomonas maltophilia Complex [Public and Environmental Health Microbiology]

The Stenotrophomonas maltophilia complex (Smc) comprises opportunistic environmental Gram-negative bacilli responsible for a variety of infections in both humans and animals. Beyond its large genetic diversity, its genetic organization in genogroups was recently confirmed through the whole-genome sequencing of human and environmental strains. As they are poorly represented in these analyses, we sequenced the whole genomes of 93 animal strains to determine their genetic background and characteristics. Combining these data with 81 newly sequenced human strains and the genomes available from RefSeq, we performed a genomic analysis that included 375 nonduplicated genomes with various origins (animal, 104; human, 226; environment, 30; unknown, 15). Phylogenetic analysis and clustering based on genome-wide average nucleotide identity confirmed and specified the genetic organization of Smc in at least 20 genogroups. Two new genogroups were identified, and two previously described groups were further divided into two subgroups each. Comparing the strains isolated from different host types and their genogroup affiliation, we observed a clear disequilibrium in certain groups. Surprisingly, some antimicrobial resistance genes, integrons, and/or clusters of attC sites lacking integron-integrase (CALIN) sequences targeting antimicrobial compounds extensively used in animals were mainly identified in animal strains. We also identified genes commonly found in animal strains coding for efflux systems. The result of a large whole-genome analysis performed by us supports the hypothesis of the putative contribution of animals as a reservoir of Stenotrophomonas maltophilia complex strains and/or resistance genes for strains in humans.

IMPORTANCE Given its naturally large antimicrobial resistance profile, the Stenotrophomonas maltophilia complex (Smc) is a set of emerging pathogens of immunosuppressed and cystic fibrosis patients. As it is group of environmental microorganisms, this adaptation to humans is an opportunity to understand the genetic and metabolic selective mechanisms involved in this process. The previously reported genomic organization was incomplete, as data from animal strains were underrepresented. We added the missing piece of the puzzle with whole-genome sequencing of 93 strains of animal origin. Beyond describing the phylogenetic organization, we confirmed the genetic diversity of the Smc, which could not be estimated through routine phenotype- or matrix-assisted laser desorption ionization–time of flight mass spectrometry (MALDI-TOF)-based laboratory tests. Animals strains seem to play a key role in the diversity of Smc and could act as a reservoir for mobile resistance genes. Some genogroups seem to be associated with particular hosts; the genetic support of this association and the role of the determinants/corresponding genes need to be explored.

Multifunctional Acidocin 4356 Combats Pseudomonas aeruginosa through Membrane Perturbation and Virulence Attenuation: Experimental Results Confirm Molecular Dynamics Simulation [Biotechnology]

A longstanding awareness in generating resistance to common antimicrobial therapies by Gram-negative bacteria has made them a major threat to global health. The application of antimicrobial peptides as a therapeutic agent would be a great opportunity to combat bacterial diseases. Here, we introduce a new antimicrobial peptide (~8.3 kDa) from probiotic strain Lactobacillus acidophilus ATCC 4356, designated acidocin 4356 (ACD). This multifunctional peptide exerts its anti-infective ability against Pseudomonas aeruginosa through an inhibitory action on virulence factors, bacterial killing, and biofilm degradation. Reliable performance over tough physiological conditions and low hemolytic activity confirmed a new hope for the therapeutic setting. Antibacterial kinetic studies using flow cytometry technique showed that the ACD activity is related to the change in permeability of the membrane. The results obtained from molecular dynamic (MD) simulation were perfectly suited to the experimental data of ACD behavior. The structure-function relationship of this natural compound, along with the results of transmission electron microscopy analysis and MD simulation, confirmed the ability of the ACD aimed at enhancing bacterial membrane perturbation. The peptide was effective in the treatment of P. aeruginosa infection in mouse model. The results support the therapeutic potential of ACD for the treatment of Pseudomonas infections.

IMPORTANCE Multidrug-resistant bacteria are a major threat to global health, and the Pseudomonas bacterium with the ability to form biofilms is considered one of the main causative agents of nosocomial infections. Traditional antibiotics have failed because of increased resistance. Thus, finding new biocompatible antibacterial drugs is essential. Antimicrobial peptides are produced by various organisms as a natural defense mechanism against pathogens, inspiring the possible design of the next generation of antibiotics. In this study, a new antimicrobial peptide was isolated from Lactobacillus acidophilus ATCC 4356, counteracting both biofilm and planktonic cells of Pseudomonas aeruginosa. A detailed investigation was then conducted concerning the functional mechanism of this peptide by using fluorescence techniques, electron microscopy, and in silico methods. The antibacterial and antibiofilm properties of this peptide may be important in the treatment of Pseudomonas infections.

A Rich Sci-fi Story

Bright Memory: Infinite is set in a sprawling, futuristic metropolis in the year 2036.

The player will take on the role of Shelia, a member of the Supernatural Science Research Organization (SRO), an organization in possession of more advanced science and technology than even the military. Bright Memory: Infinite is full of breathtaking fun and exciting action, such as navigating a spaceship that can travel through time, battling ancient samurai displaced from 1000 years ago to the present, and exploring a wormhole that has appeared on Earth.

High-Speed 3D Combat

By now I’m sure everyone has seen Bright Memory: Infinite‘s appearance on Inside Xbox. In addition to the deep and rich story, another main feature of the game is its adrenaline-pumping gameplay design. With the way I’ve designed the battle system, players can experience lightning-fast combat using a variety of skills to create their own personalized style of fighting. Basically, any skills can be combined and will generate different effects when utilized and it’s up to the player to figure out how to best use these skills both in and out of battle. Some skills can be used to overcome certain obstacles or solve certain puzzles, be it in combat or while exploring the futuristic world.

Stunning Visual Effects

The quality of the graphics of Bright Memory: Infinite for the Xbox Series X is the aspect I focused most on throughout the game’s development, in order to truly harness and show off the abilities of the next-gen system. This can particularly be seen in the wormholes appearing in the game. I’ve also made use of the dramatic visual effects made possible by the Xbox Series X, which can be seen in dynamic scenes in which wind, rain, leaves, and pebbles are visibly sucked toward a wormhole as the player explores their surroundings. These stunning visual effects are all possible thanks to DirectX Raytracing (DXR) on the Xbox Series X.

Infinite: Beyond The Mind (NS)

In a gaming era rife with flashy, bombastic AAA software, there’s definitely still a place for classic button-mashing brawlers. Ditching the cinematic bells and whistles, these are games that get straight to the point, pummelling you with pure gameplay and near-constant action. Developer Emilie Coyo and Blowfish Studios superbly tap into that late-80s gaming feel with their thrilling action romp, Infinite: Beyond the Mind

Along with the recent renaissance of Streets of Rage, games like this remind one just how appealing this forgotten genre once was. Comparisons to the old TMNT games, Final Fight, and Double Dragon tend to be dredged up soon after being dropped into the action. At the same time, the game adds a bit of depth with some platforming level design to keep you on your toes. A few imperfections and simplicities hold it back from reaching the lofty heights of those gems, to be sure. Regardless, Infinite mostly succeeds in emulating retro beat ‘em ups at their peak.

As you might expect with a 3 hour, pixelated button-mashing sprint, the story is marginal and cliche at best. You play as one of two women who have discovered the ability to use special powers. This has made them the target of a tyrannical Kingdom and its sorcerer queen, Evangelyn. The queen sends forth her brigades of pesky, heat-packing troops, bots, and artillery, which you must dash and slash your way through. 

These enemies range in terms of behaviors, as well as offensive and defensive prowess. Most, however, take the form of fairly stock military men wielding various weapons running the gamut from pistols to machine guns to grenades. There’s also a boss fight waiting for you after each multi-level biome, which proves exciting without being ridiculously tough.

Playing as either Tanya or Olga, you’ll be given mostly the same palette of moves - and it's a limited one, to be sure. Your basic jump can be extended into a double jump, which you’ll sometimes need in order to hop across some tough-to-reach platforms. Additionally, you can perform a dash move, which drains a meter displayed beneath your health bar. Dashing is fun, satisfying, and also quite useful. You can use it to dodge mid-air projectiles, evade tough spots, leap across vast gaps, and flash through deadly lasers. You’re also able to roll, which is akin to a ground-level version of this dash. 

Your offensive abilities are a bit more limited, as they basically amount to frequent close-range slashing. However, with a full meter, you’ll eventually unlock more powerful, bullet-deflecting slashes while airborne. You can bust out an explosive, screen-wiping special move a couple of times per stage, too. Given the rarity of upgrades, the speed of your character, and lack of offensive moves, Infinite: Beyond the Mind tends to stress speed and survival over sheer firepower. Wailing on tons of foes in a short time will occasionally net you extra lives, and you’re rewarded with higher scores when knocking out a stage quickly.

The majority of baddies can be slain with your close-range slash move in just a couple of hits. Still, you’ll usually want to measure their behaviors, rather than marching in while mindlessly button mashing. These anticipatory moves will help you dodge their projectiles and live to fight another day. Though you’ll stumble upon health and extra life pickups at times, these are scarce - as are the checkpoints, which only kick in at the conclusion of each “area.” 

The game further adheres to this unforgivable NES style by allowing you to save only at the end of each multi-area stage, granting you only a few lives and a short health bar in the process. This grind-it-out showdown is even tougher with the “expert” difficulty setting, as your health bar is even shorter and the ability to save is stripped away. Basically, you’ll want to be sharp, especially on the highest difficulty.

As you power through the 16 stages (each of which contains 2 or 3 bite-sized areas plus a boss), more competent and dangerous foes start showing up. You’ll come upon certain enemies covering in trenches, popping out of trucks, or posted up on high platforms. You’ll even run into tough vehicles and machinery like helicopters and robots. It certainly adds to the difficulty, while keeping feelings of repetition at bay.

One neat little inclusion is the ability to occasionally match this firepower by way of saddling up in a stationary tank. While inside, you can mow down waves of incoming troops all around you for a short time. Similarly, you’ll also be forced into static survival portions sans the tank, which can amp up the intensity quite a bit. There are even a couple of flying and scrolling areas to mix things up.

In general, the game walks that tightrope nicely in terms of difficulty - at least on its default setting. It can seem a bit chaotic at first with the breadth of projectiles from multiple areas, especially since you can only answer with close-range hits. Still, Infinite becomes far simpler once you’ve taken your lumps a couple of times and get a feel for the baddies and the stage layout. It’s easier when you’re prepared to handle that incoming tank or chopper, or can anticipate that electric wall or falling platform. Rarely did I feel like my deaths were anything but my own fault. With that said, the Double Dragon-esque push backs that sometimes knocked me into pits didn’t exactly feel just.

The control method makes a difference in tightening things up and offering a smoother experience. I wouldn’t typically stress opting for a D-pad-laced Joy-con, but in this case the improvement is vast. Seriously, if you’re inclined to make the most out of the game, consider shelling out for the HORI D-Pad left Joy-con. Infinite is still suitable when mashing the standard directional buttons, for the record, but there’s a level of swiftness and precision with the D-pad controller that meshes perfectly with the fast-paced, NES-style experience. Tanya/Olga seem to genuinely move around quicker and more sharply.

Another addition that’s both helpful and enjoyable is simultaneous co-op. In true Final Fight fashion, two players can kick some butt at once, doubling both the fun and efficiency in combat. Playing with separate Joy-cons isn’t always the most comfortable or intuitive, but the added offense makes up for some of that.

Any real blemishes of this game mostly boil down to nitpicks, and major flaws are few and far between. There’s the aforementioned prominence of push-backs while getting hit, but only rarely do these coax catastrophic death plummets. Also, while the environmental detail and layering is nice, it can sometimes be tough to make out just where you can jump and where you can’t. Jumping onto ladders to climb can also be a somewhat spotty, finicky endeavor, though these aren’t abundant at least.

Infinite: Beyond the Mind won’t blow any minds with its (literal) straightforward gameplay and brevity. Nonetheless, it makes for a fun, delightful throwback to the side-scrolling brawlers of decades past. This tight, undiluted action gameplay is complemented by a charming pixel-art-meets-anime style and appealing synthetic tunes. The 16-bit sprites add some retro character, though they’re really put to good use with the multi-layered backgrounds and colorful, vibrant biomes. These range from military bases to industrial furnaces to ice tundras. 

Overall, Infinite is a captivating action sidescroller that early 90s brawler fans, in particular, should get a kick out of. It’s a short and fairly basic experience, but worth checking out at just 10USD. Play with a friend or use a D-pad attachment to really get the most out of this already enjoyable romp.
