ncis Mycobacteria excise DNA damage in 12- or 13-nucleotide-long oligomers by prokaryotic-type dual incisions and performs transcription-coupled repair [Genomics and Proteomics] By www.jbc.org Published On :: 2020-12-11T00:06:21-08:00 In nucleotide excision repair, bulky DNA lesions such as UV-induced cyclobutane pyrimidine dimers are removed from the genome by concerted dual incisions bracketing the lesion, followed by gap filling and ligation. So far, two dual-incision patterns have been discovered: the prokaryotic type, which removes the damage in 11–13-nucleotide-long oligomers, and the eukaryotic type, which removes the damage in 24–32-nucleotide-long oligomers. However, a recent study reported that the UvrC protein of Mycobacterium tuberculosis removes damage in a manner analogous to yeast and humans in a 25-mer oligonucleotide arising from incisions at 15 nt from the 3´ end and 9 nt from the 5´ end flanking the damage. To test this model, we used the in vivo excision assay and the excision repair sequencing genome-wide repair mapping method developed in our laboratory to determine the repair pattern and genome-wide repair map of Mycobacterium smegmatis. We find that M. smegmatis, which possesses homologs of the Escherichia coli uvrA, uvrB, and uvrC genes, removes cyclobutane pyrimidine dimers from the genome in a manner identical to the prokaryotic pattern by incising 7 nt 5´ and 3 or 4 nt 3´ to the photoproduct, and performs transcription-coupled repair in a manner similar to E. coli. Photo: Chatham House. Pope Francis’ visit to Mozambique on 4–6 September comes at a critical political moment. The theme for the papal Africa trip (which also includes Madagascar and Mauritius) is ‘pilgrim of hope, peace and reconciliation’. This is especially relevant for Mozambique, as this is the first week of the official campaign for Mozambique’s sixth national elections on 15 October. It is also the one-month anniversary of the Maputo Accords for Peace and Reconciliation between the government and the armed opposition, RENAMO (and the fifth anniversary of the previous such agreement in 2014). What is unusual is that the pope accepted to visit Mozambique just after a peace accord and in the run-up to national elections. Something similar has happened only once, when Pope John Paul II visited Angola in June 1992 (following the Bicesse Accords) prior to the country’s first ever national elections in September. Unfortunately Pope John Paul’s preaching of reconciliation and pluralism failed and civil war resumed some months later, following rejection of the preliminary election results. Angola’s civil war only finally ended a decade later in 2002. The last papal visit to Mozambique was also by Pope John Paul II in 1988, when civil war was still ongoing, and the country was still a single party state. Despite the war, massive congregations attended and RENAMO reached local ceasefires and agreements to maintain electricity supply to honour the visit. Some of the seeds for the Rome peace process were laid during this trip – especially as it also represented a formal reconciliation of FRELIMO, the ruling party, with the Catholic Church. This papal visit to Mozambique is equally anticipated, as was highlighted several times during speeches at the 6 August peace agreement signing in Maputo. When I was in Maputo last month, sales of papal-pictured capulanas (a Mozambican sarong) were brisk and Mozambican television carried countdown clocks on many programmes for the touchdown of Pope Francis on national soil. The Catholic Church has played an instrumental role in promoting peace in Mozambique over the years. The 1977–92 civil war ended through negotiations hosted at the Sant’ Egidio lay community in Rome, and the current Archbishop of Bologna, Dom Matteo Zuppi (who led the Sant’ Egido negotiations in 1992 and is soon to be made a cardinal) was an official witness to 6 August accords signing. When targeted armed conflict resumed in 2013, faith groups once more re-engaged and in 2016 Sant’ Egidio once more co-led mediation efforts, less successfully than in 1991–92. Sant’ Egidio (including during a presidential visit to Rome in July) contributed to convincing the Vatican that this papal visit should occur before the October elections. President Filipe Nyusi anxiously wanted this visit to occur before the elections. He is seeking re-election for his second and final term and a papal visit should help win some votes. His party, FRELIMO, is also worried about securing a majority in the national assembly, as it has been weakened by patchy delivery of services and ongoing high-level corruption scandals. This year, President Nyusi’s priorities have been to show that he can attract international investment (such as Andarko’s recently announced final investment decision on its gas project), a peace agreement with RENAMO (the August agreements) and a papal visit, so a successful trip would complete his goals. The pope’s ‘hope, peace and reconciliation’ message of his visit is important. Twice previously, the FRELIMO-led government and RENAMO have reached definitive agreements, in Rome (1992) and Maputo (2014), but failed to fully end bloodshed. This new August 2019 agreement is the third attempt, and if it is to last, it will require political goodwill, compromise and an acceptance of more inclusive national politics by both parties. There are two immediate threats to this agreement. The first is the forthcoming 15 October elections and their conduct could make or break it. Accepting reconciliation and greater pluralism underpins this agreement, but RENAMO expects to increase its share of the parliamentary vote and win a majority in some provinces (and therefore indirectly elect their choice for governor). A second threat is the ‘Military Junta’, a RENAMO splinter group that claims to be 500 strong, but probably accounts for 80 armed persons. It rejects the 6 August agreement and warns that it could disrupt the elections. This group has asked for mediation, and hopefully can be accommodated in a side deal to the main one agreed in August, which already provides for the reintegration of over 5,000 RENAMO supporters and combatants. A recent Chatham House research paper on elite bargains in Mozambique concluded that the October elections will be the first immediate test of the August agreement. If the elections pass without significant electoral manipulation or violence and this August deal sticks on the third attempt, the domestic focus should then move onto poverty reduction, combating inequality, education and solving the new security crisis with Islamic militants in Cabo Delgado province. The pontiff received antibiotics administered intravenously during his stay, the Vatican said. In a sign of his improved health, the Vatican released details of Francis' Holy Week schedule. It said he would preside at this weekend's Palm Sunday Mass and at Easter Mass on April 9, both held in St. Peter's Square and expected to draw tens of thousands of faithful. A Vatican cardinal will be at the altar to celebrate both Masses, a recent practice due to the pontiff having a troublesome knee issue. But Francis is scheduled to celebrate Holy Thursday Mass, which this year will be held in a juvenile prison in Rome. Still unclear was whether he would attend the late-night, torch-lit Way of the Cross procession at Rome's Colosseum to mark Good Friday. Before departing Gemelli Polyclinic late Saturday morning, Francis comforted a Rome couple whose 5-year-old daughter died Friday night at the Catholic hospital. Outside, Serena Subania, mother of Angelica, sobbed as she pressed her head into the chest of the pope, who held her close and whispered words of comfort. Francis seemed eager to linger with well-wishers. When a boy showed him his arm cast, the pope made a gesture as if to ask, “Do you have a pen?” Three papal aides whipped out theirs. Francis took one of the pens and added his signature to the child's already well-autographed cast. Asked how he felt now, Francis joked, “Still alive, you know.” He gave a thumbs-up sign. Francis exited the hospital from a side entrance, but his car stopped in front of the main entrance, where a gaggle of journalists waited. He opened the car door himself and got out from the front passenger seat. Francis had a cane ready to lean on. After chatting, he got back into the white Fiat 500 car that drove him away from Gemelli Polyclinic. But instead of heading straight home, his motorcade sped right past Vatican City and went to St. Mary Major Basilica, a Rome landmark that is one of his favorites. There, startled tourists rushed to snap photos of him as he sat in a wheelchair, which he has used often to navigate longer distances in recent years due to a chronic knee problem. When he emerged after praying, residents and tourists in the street called out repeatedly, “Long live the pope!” and clapped. Francis spent 10 days at the same hospital in July 2021 following intestinal surgery for a bowel narrowing, After his release back then, he also stopped to offer prayers of thanksgiving at St. Mary Major Basilica, which is home to an icon depicting the Virgin Mary. He also visits the church upon returning from trips abroad. Before leaving the hospital Saturday, Francis, while chatting with journalists, praised medical workers, saying they "show great tenderness." “We sick are capricious. Francis stopped to talk to reporters again before he was driven into the Vatican through a gate of the tiny walled city-state, where he lives at a Holy See hotel. Mexico however is a country where journalists sometimes do their work under threats to themselves and their families and going to the police is not always a good idea. Sara soon finds that if she wants to pursue this particular story she could be putting the lives of herself, her younger brother Emiiano and their mother in danger.Emiliano is a soccer star at his high school and in addition to this he is also a member of a school group called Jiparis who do hikes through the desert in order to build character in the young men, some of whom were involved in unsavory activities before joining the group.One part of the Jipari pledge goes , "I will be honest with myself and others". This is easier said than done especially in a city like Juarez. One of the characters tells Emiliano, "everything is a spiderweb" and the speed with which he is enveloped in said web is astounding. Emiliano tells himself that he wants to help his family and friends out but are those his real reasons? Like any teen he wants to be seen as cool and there is also the small matter of a girl he wants to impress.The first part of the book is told from each character's viewpoint and the author weaves the tale together in a very credible way showing how circumstancesMake no mistake, Disappeared is not a peaches and cream, hunky dory teen novel. It is a gritty and very realistic novel with a ripped from the headlines quality to it. The city of Juarez and the violence there doesn't dominate the headlines as it did a few years ago, but Stork's novel is a timely reminder that evil still exists and that it takes many people working in tandem to defeat it. Steve Julian: Business analyst Mark Lacter, that might come in handy given how crowded California's air corridor has become... Mark Lacter: We'll talk about the hyper-loop in a moment, Steve, but yes, the L.A.-to-San Francisco air route is the busiest in the U.S., and it's already the most competitive. We're talking about more than 50 flights a day, which - if you spread them out between six in the morning and 10:30 at night - there'd be one flight every 20 minutes. But, Delta obviously thinks there's room for more because it's announced an hourly shuttle between the two cities. That's another 14 daily flights beginning September 3. The airline will be using a somewhat smaller jet, and it sounds as if the focus will be on the business traveler, with free newspapers, wine, and beer. Julian: How much will it cost, do we know? Lacter: As usual, it's a lot cheaper if you make an advance purchase, but if you're buying your tickets at the last minute - which is what a lot of business travelers do - roundtrip runs a hefty $430. Actually, this Bay Area shuttle is just the latest effort by Delta to expand out of LAX, which is different from other major airports in that it doesn't have any one airline that dominates (United has a slight edge in market share over American, with Delta about three percentage points behind). American also has been adding flights out of LAX. Julian: Sounds like the airline business is improving... Lacter: That's what happens when you pack planes to the absolute max, which is bad news for travelers being crammed into coach seats. But it's good news for LAX, which continues to be the airport of choice among airlines looking to add service - matter of fact, domestic passenger traffic was up almost 8 percent in June compared with a year earlier. Some of those gains might be at the expense of service elsewhere - most especially Ontario Airport, which has seen a big exodus among airlines and passengers. Ontario city officials have been trying to regain control of the airport, which has been operated by the city of Los Angeles. Julian: Back to the hyper-loop - is this kind of transport possible? Lacter: Well, it's the brainchild of billionaire Elon Musk, and you never say never with this guy. He started the electric car company Tesla and the private space company Space X. The hyper-loop is a high-speed system of passenger pods that would travel on a cushion of air (think of air hockey table). The pods would travel at more than 700 miles per hour, but they wouldn't result in sonic booms that severely restricted the Concorde aircraft. Of course, anything that promises super-speed travel is bound to get people talking - and, from what the physics professors are saying, the Musk idea seems feasible. Julian: How would its cost compare to the bullet train? Lacter: He says a lot cheaper. The price tag on the train is $70 billion at last check; Musk says he can do his for $6 billion. But, the issue isn't so much the cost or even the technology, but the politics. As a rule, governments do not think outside the box, and that's what a project like this is all about. Already, you have bullet train supporters saying that the hyper-loop is impossible, but what they're really saying is we have a lot riding on the train, and we don't want this guy to mess it up. Julian: But, how much demand is there for high-speed transport? Lacter: You'd think there would be a lot, but when Boeing came up with a nifty idea for a souped-up plane that would shave almost an hour from L.A. to New York, the airlines said no because it would require more fuel - and that would mean raising fares. Musk says his system would be a lot cheaper than traveling by plane, which could be a game changer in the attitudes about going places. But, those attitudes won't change until the thing is actually built, and that can't realistically happen until attitudes change. That's the ultimate problem.
