Catawba Co. joins other area counties & cities to approve settlement of Catawba River Basin Inter Basin Transfer issue

The Catawba County Board of Commissioners, at its January 19, 2010 meeting, joined with other local governments across the region in approving the agreement. The main points of the agreement hinged on Concord and Kannapolis modifying their ability, contained in their IBT certificate, to withdraw 10 million gallons of water per day (MGD) from the Catawba River at all times, by significantly limiting withdrawals during times of drought. The agreement limits withdrawals to 6 MGD during times of most severe drought, or �exceptional� drought; 7 MGD during �extreme� drought; 8.5 MGD during �severe� drought; and 9 MGD during �moderate� drought. Further, the agreement restricts Concord and Kannapolis from withdrawing more than 3 MGD from the Catawba until July 1, 2015, and after they first are withdrawing 5 MGD from the Yadkin River.

China’s New Cross-Border Data Transfer Rules Substantially Reduce Compliance Burdens for Multinational Employers

Multinational employers operating in China have been waiting since September 2023 for the Cyberspace Administration of China (CAC) to finalize proposed revisions to its complex and burdensome rules for cross-border data transfers.  Relief arrived on March 22, 2024, when the CAC published the “Provisions on Promoting and Regulating Cross-border Data Flows” (the “Approved Provisions”), which went into effect on the same day.

File Transfer Solutions Targeted by CL0P Ransomware Group

The CL0P Ransomware Group, also known as TA505, has exploited zero-day vulnerabilities across a series of file transfer solutions since December 2020. File transfer solutions often contain sensitive information from a variety of organizations. This stolen information is used to extort victims to pay ransom demands. In 2023, CL0P claimed credit for the exploitation of vulnerabilities in both Fortra’s GoAnywhere Managed File Transfer (MFT) and Progress Software’s MOVEit Transfer solutions.

Research conducted as part of security audits has revealed additional vulnerabilities. This dashboard contains a list of vulnerabilities known to be exploited by the CL0P ransomware group as well as other vulnerabilities that could be leveraged by CL0P and other threat actors. Operations teams can use this data to identify the assets affected by the associated CVEs targeted by the CL0P ransomware group. The following Nessus plugins identify the affected vulnerabilities:

OSCE supports roundtable discussion on introduction of Pollutant Release and Transfer Register in Kazakhstan

ASTANA, 14 July 2016 – The introduction of the Pollutant Release and Transfer Register (PRTR) in Kazakhstan was the focus of a roundtable discussion organized in Astana today by the OSCE Programme Office in Astana in partnership with the Energy Ministry’s Information and Analytical Centre of Environment Protection and the UN Development Programme in Kazakhstan.

Some 80 representatives of the Aarhus Centres, regional and central government entities, environmental NGOs and private sector focused on recent changes in national environmental legislation, guiding rules in implementing PRTR, international best practices and the role of the Aarhus Centres in promoting PRTR in the regions.

Joldasov Zulfuhar, Deputy Chairperson of the Committee of Environmental Regulation, Control and State Inspection in the oil and gas sector of Kazakhstan’s Energy Ministry, said:  “Taking into account that environmental damage impacts all spheres of life, a state register of pollutant release and transfer is particularly relevant. We are convinced of the need to co-operate with all interested parties and consolidate our efforts in addressing the emerging issues in the field of environmental protection and sustainable development.”

Mirco Guenther, Deputy Head of the OSCE Programme Office in Astana, said: "PRTR management can lead to a significant reduction in environmental and social risks to promote a constructive dialogue between interested parties, as well as to achieve a balance of interests and protection of environmental rights of citizens. The OSCE Programme Office in Astana stands ready to support Kazakhstan’s initiative to comply fully with all their obligations under the Aarhus Convention."

The PRTR Protocol was adopted in 2003, in Kyiv, Ukraine and is a legally binding instrument that requires parties to establish publicly accessible registers containing information on the release and transfer of pollutants.

The event is part of the Programme Office’s longstanding efforts in promoting the international standards of the Aarhus Convention in Kazakhstan.

Officials say the United States has given its approval for the Netherlands and Denmark to deliver F-16s to Ukraine. The defense ministers of those two NATO countries announced the decision Friday. It is a major gain for Kyiv even though the fighter jets won’t have an impact any time soon on the almost 18-month war. It was not immediately clear when the first F-16s might enter the conflict. Denmark says it will hand over some of its F-16s only after receiving its new F-35 jet fighters, which are due to start arriving on Oct. 1. Officials have previously said that Ukrainian pilots will need six to eight months of training on the F-16s.

Genetic evidence for partial redundancy between the arginine methyltransferases CARM1 and PRMT6 [Signal Transduction]

CARM1 is a protein arginine methyltransferase (PRMT) that acts as a coactivator in a number of transcriptional programs. CARM1 orchestrates this coactivator activity in part by depositing the H3R17me2a histone mark in the vicinity of gene promoters that it regulates. However, the gross levels of H3R17me2a in CARM1 KO mice did not significantly decrease, indicating that other PRMT(s) may compensate for this loss. We thus performed a screen of type I PRMTs, which revealed that PRMT6 can also deposit the H3R17me2a mark in vitro. CARM1 knockout mice are perinatally lethal and display a reduced fetal size, whereas PRMT6 null mice are viable, which permits the generation of double knockouts. Embryos that are null for both CARM1 and PRMT6 are noticeably smaller than CARM1 null embryos, providing in vivo evidence of redundancy. Mouse embryonic fibroblasts (MEFs) from the double knockout embryos display an absence of the H3R17me2a mark during mitosis and increased signs of DNA damage. Moreover, using the combination of CARM1 and PRMT6 inhibitors suppresses the cell proliferation of WT MEFs, suggesting a synergistic effect between CARM1 and PRMT6 inhibitions. These studies provide direct evidence that PRMT6 also deposits the H3R17me2a mark and acts redundantly with CARM1.

Inhibition of the SUV4-20 H1 histone methyltransferase increases frataxin expression in Friedreich's ataxia patient cells [Gene Regulation]

The molecular mechanisms of reduced frataxin (FXN) expression in Friedreich's ataxia (FRDA) are linked to epigenetic modification of the FXN locus caused by the disease-associated GAA expansion. Here, we identify that SUV4-20 histone methyltransferases, specifically SUV4-20 H1, play an important role in the regulation of FXN expression and represent a novel therapeutic target. Using a human FXN–GAA–Luciferase repeat expansion genomic DNA reporter model of FRDA, we screened the Structural Genomics Consortium epigenetic probe collection. We found that pharmacological inhibition of the SUV4-20 methyltransferases by the tool compound A-196 increased the expression of FXN by ∼1.5-fold in the reporter cell line. In several FRDA cell lines and patient-derived primary peripheral blood mononuclear cells, A-196 increased FXN expression by up to 2-fold, an effect not seen in WT cells. SUV4-20 inhibition was accompanied by a reduction in H4K20me2 and H4K20me3 and an increase in H4K20me1, but only modest (1.4–7.8%) perturbation in genome-wide expression was observed. Finally, based on the structural activity relationship and crystal structure of A-196, novel small molecule A-196 analogs were synthesized and shown to give a 20-fold increase in potency for increasing FXN expression. Overall, our results suggest that histone methylation is important in the regulation of FXN expression and highlight SUV4-20 H1 as a potential novel therapeutic target for FRDA.

Characterizing human {alpha}-1,6-fucosyltransferase (FUT8) substrate specificity and structural similarities with related fucosyltransferases [Protein Structure and Folding]

Mammalian Asn-linked glycans are extensively processed as they transit the secretory pathway to generate diverse glycans on cell surface and secreted glycoproteins. Additional modification of the glycan core by α-1,6-fucose addition to the innermost GlcNAc residue (core fucosylation) is catalyzed by an α-1,6-fucosyltransferase (FUT8). The importance of core fucosylation can be seen in the complex pathological phenotypes of FUT8 null mice, which display defects in cellular signaling, development, and subsequent neonatal lethality. Elevated core fucosylation has also been identified in several human cancers. However, the structural basis for FUT8 substrate specificity remains unknown.Here, using various crystal structures of FUT8 in complex with a donor substrate analog, and with four distinct glycan acceptors, we identify the molecular basis for FUT8 specificity and activity. The ordering of three active site loops corresponds to an increased occupancy for bound GDP, suggesting an induced-fit folding of the donor-binding subsite. Structures of the various acceptor complexes were compared with kinetic data on FUT8 active site mutants and with specificity data from a library of glycan acceptors to reveal how binding site complementarity and steric hindrance can tune substrate affinity. The FUT8 structure was also compared with other known fucosyltransferases to identify conserved and divergent structural features for donor and acceptor recognition and catalysis. These data provide insights into the evolution of modular templates for donor and acceptor recognition among GT-B fold glycosyltransferases in the synthesis of diverse glycan structures in biological systems.

Hydrogen/deuterium exchange memory NMR reveals structural epitopes involved in IgE cross-reactivity of allergenic lipid transfer proteins [Protein Structure and Folding]

Identification of antibody-binding epitopes is crucial to understand immunological mechanisms. It is of particular interest for allergenic proteins with high cross-reactivity as observed in the lipid transfer protein (LTP) syndrome, which is characterized by severe allergic reactions. Art v 3, a pollen LTP from mugwort, is frequently involved in this cross-reactivity, but no antibody-binding epitopes have been determined so far. To reveal human IgE-binding regions of Art v 3, we produced three murine high-affinity mAbs, which showed 70–90% coverage of the allergenic epitopes from mugwort pollen–allergic patients. As reliable methods to determine structural epitopes with tightly interacting intact antibodies under native conditions are lacking, we developed a straightforward NMR approach termed hydrogen/deuterium exchange memory (HDXMEM). It relies on the slow exchange between the invisible antigen-mAb complex and the free 15N-labeled antigen whose 1H-15N correlations are detected. Due to a memory effect, changes of NH protection during antibody binding are measured. Differences in H/D exchange rates and analyses of mAb reactivity to homologous LTPs revealed three structural epitopes: two partially cross-reactive regions around α-helices 2 and 4 as well as a novel Art v 3–specific epitope at the C terminus. Protein variants with exchanged epitope residues confirmed the antibody-binding sites and revealed strongly reduced IgE reactivity. Using the novel HDXMEM for NMR epitope mapping allowed identification of the first structural epitopes of an allergenic pollen LTP. This knowledge enables improved cross-reactivity prediction for patients suffering from LTP allergy and facilitates design of therapeutics.
