Integration of Swimming-Related Synaptic Excitation and Inhibition by olig2+ Eurydendroid Neurons in Larval Zebrafish Cerebellum

The cerebellum influences motor control through Purkinje target neurons, which transmit cerebellar output. Such output is required, for instance, for larval zebrafish to learn conditioned fictive swimming. The output cells, called eurydendroid neurons (ENs) in teleost fish, are inhibited by Purkinje cells and excited by parallel fibers. Here, we investigated the electrophysiological properties of glutamatergic ENs labeled by the transcription factor olig2. Action potential firing and synaptic responses were recorded in current clamp and voltage clamp from olig2+ neurons in immobilized larval zebrafish (before sexual differentiation) and were correlated with motor behavior by simultaneous recording of fictive swimming. In the absence of swimming, olig2+ ENs had basal firing rates near 8 spikes/s, and EPSCs and IPSCs were evident. Comparing Purkinje firing rates and eurydendroid IPSC rates indicated that 1-3 Purkinje cells converge onto each EN. Optogenetically suppressing Purkinje simple spikes, while preserving complex spikes, suggested that eurydendroid IPSC size depended on presynaptic spike duration rather than amplitude. During swimming, EPSC and IPSC rates increased. Total excitatory and inhibitory currents during sensory-evoked swimming were both more than double those during spontaneous swimming. During both spontaneous and sensory-evoked swimming, the total inhibitory current was more than threefold larger than the excitatory current. Firing rates of ENs nevertheless increased, suggesting that the relative timing of IPSCs and EPSCs may permit excitation to drive additional eurydendroid spikes. The data indicate that olig2+ cells are ENs whose activity is modulated with locomotion, suiting them to participate in sensorimotor integration associated with cerebellum-dependent learning.

SIGNIFICANCE STATEMENT The cerebellum contributes to movements through signals generated by cerebellar output neurons, called eurydendroid neurons (ENs) in fish (cerebellar nuclei in mammals). ENs receive sensory and motor signals from excitatory parallel fibers and inhibitory Purkinje cells. Here, we report electrophysiological recordings from ENs of larval zebrafish that directly illustrate how synaptic inhibition and excitation are integrated by cerebellar output neurons in association with motor behavior. The results demonstrate that inhibitory and excitatory drive both increase during fictive swimming, but inhibition greatly exceeds excitation. Firing rates nevertheless increase, providing evidence that synaptic integration promotes cerebellar output during locomotion. The data offer a basis for comparing aspects of cerebellar coding that are conserved and that diverge across vertebrates.

Whale sharks increase swimming effort while filter feeding, but appear to maintain high foraging efficiencies [RESEARCH ARTICLE]

David E. Cade, J. Jacob Levenson, Robert Cooper, Rafael de la Parra, D. Harry Webb, and Alistair D. M. Dove

Whale sharks (Rhincodon typus Smith 1828) – the largest extant fish species – reside in tropical environments, making them an exception to the general rule that animal size increases with latitude. How this largest fish thrives in tropical environments that promote high metabolism but support less robust zooplankton communities has not been sufficiently explained. We used open-source inertial measurement units (IMU) to log 397 hours of whale shark behavior in Yucatan, Mexico, at a site of both active feeding and intense wildlife tourism. Here we show that the strategies employed by whale sharks to compensate for the increased drag of an open mouth are similar to ram-feeders five orders of magnitude smaller and one order of magnitude larger. Presumed feeding constituted 20% of the total time budget of four sharks, with individual feeding bouts lasting up to 11 consecutive hrs. Compared to normal, sub-surface swimming, three sharks increased their stroke rate and amplitude while surface feeding, while one shark that fed at depth did not demonstrate a greatly increased energetic cost. Additionally, based on time-depth budgets, we estimate that aerial surveys of shark populations should consider including a correction factor of 3 to account for the proportion of daylight hours that sharks are not visible at the surface. With foraging bouts generally lasting several hours, interruptions to foraging during critical feeding periods may represent substantial energetic costs to these endangered species, and this study presents baseline data from which management decisions affecting tourist interactions with whale sharks may be made.

COVID-19: Radio Club staffer called to change swimming pool water, electrocuted

A swimming pool maintenance employee working at the Bombay Presidency Radio Club (BPRC) in Colaba amid the Coronavirus lockdown died of electrocution on Thursday. Ramchandra Bhuneshwar's body was seen floating in the swimming pool by another staffer.

Deputy Commissioner of Police Sangramsinh Nishandar said an Accidental Death Report has been filed. "Prima facie, a 42-year-old man in charge of maintaining the swimming pool at Colaba's Radio Club died of electrocution. We have registered a case under Section 188 (disobedience to order duly promulgated by public servant) of the Indian Penal Code against Radio Club officials and the contractor for breaking lockdown norms," he said.

Senior police inspector of Colaba police Shivaji Phadtare said Bhuneshwar was taking water out of the pool with the help of an electric pump when he got electrocuted.
Sources said that Bhuneshwar, a contractual employee, was asked by his contractor Babu Khomrekar to clean the water of the swimming pool a week ago. "At the time of conducting the panchnama, Khomrekar was called by Colaba police. He told the police that Adi Mistry, a Managing Committee Member working as the Chairman of Swimming Pool and Gym at the Radio Club, told him to get Bhuneshwar to change the pool water," a source working at the Radio Club said.

Bhuneshwar is survived by wife, Vimal, two daughters and a six-year-old son. His relative, Siddhant Gaikwad, said, "Bhuneshwar was asked to continue maintenance work at Radio Club's swimming pool even during the lockdown. He lives in Murud Janjira. When no permanent staff is working at the club, why was he forced to continue his work?"

President of Radio Club, Harish Kumar Garg, said, "I am not aware how Bhuneshwar died and what was he doing at the Radio Club amid a lockdown as I am a 72-year-old man and I have been staying home."

Ramchandra Bhuneshwar

Sources said nearly a dozen Radio Club employees living at the premises spoke to the Colaba police during the panchanama. "Committee members forced these workers to leave the club amid the lockdown because they spoke to the police," source added.

Club members speak

Advocate Ravi Goenka, a member of the club, said, "We all are following the lockdown rules and sitting at home. And the rich and powerful people who are on the committee of tony clubs (i.e Radio Club) call staff to work in spite of the lockdown."

The Radio Club committee refused to answer calls. A club member said angrily, "Calling in staff to work is exploitation and violation of the lockdown."

Several members expressed surprise that the pool was filled with water and not drained as no one swims there amid the lockdown. Another member said, "We do not want this brushed away as an Accidental Death Report. A poor employee has lost his life, let us get to the bottom of this through a thorough investigation."

Swimming world championships moved from 2021 to 2022

The next swimming world championships, scheduled for summer 2021 in Japan, have been pushed back until May 2022 following the postponement of the Olympic Games, the sport's governing body said Monday. The world championships were due to be held in Fukuoka next year from July 16-August 1 but will instead take place from May 13-29 in 2022, the international swimming federation (FINA) said in a statement.

The 2020 Olympic Games were delayed by a year due to the coronavirus pandemic, and will now run from July 23-August 8 in 2021. "After liaising with the relevant stakeholders and receiving feedback from them, we have no doubt that the decision taken will provide the best possible conditions for all participants at the championships," said FINA president Julio Maglione. "At a time of unprecedented uncertainty, FINA hopes the announcement of these dates will allow for some clarity in planning for all concerned." The world aquatics championships also feature diving, open water swimming, synchronised swimming and water polo.
