shapes The lymph node stromal laminin α5 shapes alloimmunity By www.jci.org Published On :: Lymph node stromal cells (LNSCs) regulate immunity through constructing lymphocyte niches. LNSC-produced laminin α5 (Lama5) regulates CD4+ T cells but the underlying mechanisms of its functions are poorly understood. Here we show that depleting Lama5 in LNSCs resulted in decreased Lama5 protein in the LN cortical ridge (CR) and around high endothelial venules (HEVs). Lama5 depletion affected LN structure with increased HEVs, upregulated chemokines, and cell adhesion molecules, and led to greater numbers of Tregs in the T cell zone. Mouse and human T cell transendothelial migration and T cell entry into LNs were suppressed by Lama5 through the receptors α6 integrin and α-dystroglycan. During immune responses and allograft transplantation, depleting Lama5 promoted antigen-specific CD4+ T cell entry into the CR through HEVs, suppressed T cell activation, and altered T cell differentiation to suppressive regulatory phenotypes. Enhanced allograft acceptance resulted from depleting Lama5 or blockade of T cell Lama5 receptors. Lama5 and Lama4/Lama5 ratios in allografts were associated with the rejection severity. Overall, our results demonstrated that stromal Lama5 regulated immune responses through altering LN structures and T cell behaviors. This study delineated a stromal Lama5–T cell receptor axis that can be targeted for immune tolerance modulation. Full Article
shapes {beta}-Cell Stress Shapes CTL Immune Recognition of Preproinsulin Signal Peptide by Posttranscriptional Regulation of Endoplasmic Reticulum Aminopeptidase 1 By diabetes.diabetesjournals.org Published On :: 2020-03-20T11:50:28-07:00 The signal peptide of preproinsulin is a major source for HLA class I autoantigen epitopes implicated in CD8 T cell (CTL)–mediated β-cell destruction in type 1 diabetes (T1D). Among them, the 10-mer epitope located at the C-terminal end of the signal peptide was found to be the most prevalent in patients with recent-onset T1D. While the combined action of signal peptide peptidase and endoplasmic reticulum (ER) aminopeptidase 1 (ERAP1) is required for processing of the signal peptide, the mechanisms controlling signal peptide trimming and the contribution of the T1D inflammatory milieu on these mechanisms are unknown. Here, we show in human β-cells that ER stress regulates ERAP1 gene expression at posttranscriptional level via the IRE1α/miR-17-5p axis and demonstrate that inhibition of the IRE1α activity impairs processing of preproinsulin signal peptide antigen and its recognition by specific autoreactive CTLs during inflammation. These results underscore the impact of ER stress in the increased visibility of β-cells to the immune system and position the IRE1α/miR-17 pathway as a central component in β-cell destruction processes and as a potential target for the treatment of autoimmune T1D. shapes Using Fear of the “Other,” Orbán Reshapes Migration Policy in a Hungary Built on Cultural Diversity By www.migrationpolicy.org Published On :: Wed, 09 Oct 2019 11:41:59 -0400 The recent rise in xenophobia in Hungary stands in marked contrast with the country's rich migration history. After 390,000 migrants and asylum seekers arrived in 2015, the government of Viktor Orbán issued policies to significantly limit migration and enacted a law criminalizing humanitarian assistance to migrants. This country profile examines Hungary’s migration past and present, tracing the country’s multicultural heritage to the current wave of anti-immigrant sentiment. Full Article

I have a script that creates highlight shapes on the y* drawing layer. My understand is the highlight is a virtual shape. The shapes go away when the cadence session is closed or when you close data of that cellview if it is not global. If they are vitual shapes it would be okay to use valid or process layers when I create the highlight set with geCreateHilightSet. I've tried using axlShapeAutoVoid, but this only voids the pins, not the route keepouts created by nc_backdrill. I also tried selecting the shape, individually, then running axlShapeAutoVoid. That was unsuccessful, also. planeShapes is a list of shapes I created. shapes Dealing with Delhi: How culture shapes India’s Middle East policy By webfeeds.brookings.edu Published On :: Tue, 22 Dec 2015 00:00:00 -0500 Indian Prime Minister Narendra Modi’s recent visit to the United Arab Emirates revealed New Delhi’s intention to bolster bilateral relations with the Gulf states. It was the first visit by an Indian prime minister in over 30 years, demonstrating the country’s renewed focus on expanding ties with the region it has always called “West Asia.” Although India and the Middle East share a long history of trade, immigration and cultural exchange, relations have yet to reach their full potential. Read "Dealing with Delhi: How culture shapes India’s Middle East policy" In this policy briefing, Kadira Pethiyagoda highlights the importance of an under-reported aspect of the relationship – culture. The author explains the role it plays in India’s policies toward the region, particularly under the current government, and argues that Gulf states need to understand the impact of Indian values and identity. Pethiyagoda provides recommendations on how the Gulf states can, through better understanding the cultural drivers of Indian foreign policy, build stronger ties with India, thereby advancing both economic and strategic interests. shapes How humans survived an ancient volcanic winter and how disgust shapes ecosystems By traffic.omny.fm Published On :: Thu, 15 Mar 2018 14:15:00 -0400 When Indonesia’s Mount Toba blew its top some 74,000 years ago, an apocalyptic scenario ensued: Tons of ash and debris entered the atmosphere, coating the planet in ash for 2 weeks straight and sending global temperatures plummeting. Despite the worldwide destruction, humans survived. Sarah Crespi talks with Online News Editor Catherine Matacic about how life after Toba was even possible—were humans decimated, or did they rally in the face of a suddenly extra hostile planet? Next, Julia Buck of the University of California, Santa Barbara, joins Sarah to discuss her Science commentary piece on landscapes of disgust. You may have heard of a landscape of fear—how a predator can influence an ecosystem not just by eating its prey, but also by introducing fear into the system, changing the behavior of many organisms. Buck and colleagues write about how disgust can operate in a similar way: Animals protect themselves from parasites and infection by avoiding disgusting things such as dead animals of the same species or those with disease.
