Marked and rapid effects of pharmacological HIF-2α antagonism on hypoxic ventilatory control

Hypoxia-inducible factor (HIF) is strikingly upregulated in many types of cancer, and there is great interest in applying inhibitors of HIF as anticancer therapeutics. The most advanced of these are small molecules that target the HIF-2 isoform through binding the PAS-B domain of HIF-2α. These molecules are undergoing clinical trials with promising results in renal and other cancers where HIF-2 is considered to be driving growth. Nevertheless, a central question remains as to whether such inhibitors affect physiological responses to hypoxia at relevant doses. Here, we show that pharmacological HIF-2α inhibition with PT2385, at doses similar to those reported to inhibit tumor growth, rapidly impaired ventilatory responses to hypoxia, abrogating both ventilatory acclimatization and carotid body cell proliferative responses to sustained hypoxia. Mice carrying a HIF-2α PAS-B S305M mutation that disrupts PT2385 binding, but not dimerization with HIF-1β, did not respond to PT2385, indicating that these effects are on-target. Furthermore, the finding of a hypomorphic ventilatory phenotype in untreated HIF-2α S305M mutant mice suggests a function for the HIF-2α PAS-B domain beyond heterodimerization with HIF-1β. Although PT2385 was well tolerated, the findings indicate the need for caution in patients who are dependent on hypoxic ventilatory drive.

Mature myelin maintenance requires Qki to coactivate PPARβ-RXRα–mediated lipid metabolism

Lipid-rich myelin forms electrically insulating, axon-wrapping multilayers that are essential for neural function, and mature myelin is traditionally considered metabolically inert. Surprisingly, we discovered that mature myelin lipids undergo rapid turnover, and quaking (Qki) is a major regulator of myelin lipid homeostasis. Oligodendrocyte-specific Qki depletion, without affecting oligodendrocyte survival, resulted in rapid demyelination, within 1 week, and gradually neurological deficits in adult mice. Myelin lipids, especially the monounsaturated fatty acids and very-long-chain fatty acids, were dramatically reduced by Qki depletion, whereas the major myelin proteins remained intact, and the demyelinating phenotypes of Qki-depleted mice were alleviated by a high-fat diet. Mechanistically, Qki serves as a coactivator of the PPARβ-RXRα complex, which controls the transcription of lipid-metabolism genes, particularly those involved in fatty acid desaturation and elongation. Treatment of Qki-depleted mice with PPARβ/RXR agonists significantly alleviated neurological disability and extended survival durations. Furthermore, a subset of lesions from patients with primary progressive multiple sclerosis were characterized by preferential reductions in myelin lipid contents, activities of various lipid metabolism pathways, and expression level of QKI-5 in human oligodendrocytes. Together, our results demonstrate that continuous lipid synthesis is indispensable for mature myelin maintenance and highlight an underappreciated role of lipid metabolism in demyelinating diseases.

New NFC Specification Will Let Smartphones Charge Small Devices

A new NFC specification announced this week by the NFC Froum will allow future NFC enabled devices to offer wireless charging capabilities, which means a smartphone could be used to charge a small accessory like headphones.


According to the NFC Forum, the Wireless Charging Specification (WLC) will allow smartphones or other NFC charging devices to wirelessly charge small, battery-powered consumer and IoT devices at a power transfer rate of up to one watt.

The 1W rating is much slower than the Qi-based standard used by iPhones and other smartphones. Qi-based wireless charging on the iPhone maxes out at 7.5W, but is even faster on some Android devices.

Charging over NFC would require new hardware, and it's not a feature that can be added to existing devices. The NFC Forum believes the WLC specification could be used to complement Qi-based wireless charging.

It works using a single antenna to manage communications and charging, which is convenient for low-power devices like smart watches, fitness trackers, and earbuds that already use NFC for connectivity because there's no need to build in Qi support.

"The NFC Forum's Wireless Charging Technical Specification allows for wireless charging of small battery-powered devices like those found in many of the estimated 36 billion IoT devices in use today," said Koichi Tagawa, chair, NFC Forum. "NFC wireless charging is truly transformative because it changes the way we design and interact with small, battery-powered devices as the elimination of plugs and cords enables the creation of smaller, hermetically-sealed devices."
Apple in 2015 joined the NFC Forum and participates in the approval of new NFC specifications and developments.

Rumors in 2019 suggested that Apple was working on bilateral wireless charging that would allow its iPhones to charge the AirPods and the Apple Watch, but the feature was ultimately nixed. At the time, Apple analyst Ming-Chi Kuo said that the two-way wireless charging feature Apple was exploring did not meet Apple's requirements.

Samsung has implemented bilateral wireless charging in its smartphones, but the Wireless PowerShare option does not use NFC and is powered by the Qi-based charging coils in the device. Samsung's smartphones can charge other smartphones or accessories like headphones that support Qi wireless chargers.

I'll be teaching two, full-day lighting workshops in the suburban Baltimore/DC area on December 6 and 7.

They are two separate one-day events. These are small-group workshops, with a maximum class size of 12 people each day. These are shooting workshops, and you'll be behind (and/or in front of!) a camera for most of the day.

Assuming you arrive at this class with a basic understanding of f/stops and shutter speeds, you will leave with a strong foundation in the fundamentals of off-camera lighting. You will be comfortable using single and multiple lights both alone and/or balanced with a mix of ambient lighting.

All lighting gear will be provided. We will also provide lunch.

You will need to bring a camera that is adjustable in manual mode and has a hot shoe connection for a flash, a lens that covers normal and/or portrait range, one or two fully charged batteries and an empty storage card. Maybe a notebook and pen if you like.

That's it. Just show up ready to learn and to have fun. Leave the rest to me.

Every year I dread the end of summer, but then fall comes and I think, "No, this is my favorite time of year!" Fall is apples, and pumpkins, and pea soup, and cider mills, and wineries, and music, and being cozy. 



  


This smoothie bowl doesn't contain any actual pumpkin, but will convince you otherwise!Instead, I've used carrots, which are always available. It will get your day started in a delicious way and give you plenty of energy to enjoy whatever these glorious fall days bring your way!






ingredients
  • 1 cup carrots, cut into chunks
  • 1 banana, sliced and frozen
  • 1 cup almond milk 
  • 1 teaspoon cinnamon
  • 1/2 teaspoon ginger
  • 1/4 teaspoon allspice
  • pinch nutmeg
  • 1/2 cup chopped dates
  • 1/4 cup chopped pecans or other nuts
  • 1 cup chopped apples

directions
  1. In a high powered blender, puree the carrots, banana, and almond milk until very smooth.
  2. Add in the cinnamon, ginger, allspice, and nutmeg and pulse a few times to blend well.
  3. Pour into a bowl and add the chopped dates, pecans, and apples. A sprinkle of coconut flakes goes well, too!

Pineapple Green Smoothie ~ Raw Food Vegan Smoothie Recipe



This raw and healthy pineapple party in your mouth is super simple and perfect for spring. Hawaiian pineapple season is April and May, even though they're available year round. Price and quality will be the best over the next few months so take advantage of this sweet abundance and enjoy this vegan treat for breakfast or an anytime snack.

Oh, and to make this extra thick and frosty, cube and freeze pineapple and any subsequent pineapple juice in ice cube trays. Store frozen cubes in plastic bags for ultra easy prep.



   



Pineapple Green Smoothie
serves 2 ~ $1.75 per serving

ingredients
  • 3 cups chopped romaine (or favorite green) ($1.00)
  • 1 cup cubed pineapple, frozen ($2.00)
  • 1 banana, sliced and frozen ($.20)
  • 2 tablespoons lemon juice (optional) ($.20)
  • 1 teaspoon vanilla ($.10)
  • 1 cup water
  • 10 ice cubes
  • stevia to taste (optional)
directions
  • In a high powered blender, puree the greens until liquefied and very smooth. 
  • Add the rest of the ingredients and puree until smooth.

Chocolate Orange Smoothie
serves 1 ~ $2.20 per serving 


You have to remind yourself this is good for you. It's so delicious it feels downright decadent. The hemp milk makes it super creamy and luscious.

ingredients
  • 1 orange, peeled and frozen ($.50)
  • 2 bananas, peeled, sliced, and frozen ($.40)
  • 1 cup hemp milk ($1.00)
  • 3 tablespoons cacao powder ($.30) 
  • 1/2 teaspoon orange extract
  • few drops stevia (optional)
  • pinch salt
  • 6 ice cubes
  • water for blending, if necessary

directions
  1. In a high speed blender, puree all ingredients until very smooth and creamy. 
  2. Add water if needed for ease of blender or if a thinner smoothie is desired.
