DigiKoo: A German Solution to the Utility Data Sharing Conundrum

For most of their history, in North America, electrical utilities have been centralized distribution networks. Utility operated generation resources are the hub of the network and electricity flows one-way via distribution networks largely controlled by the same utilities. In this model, there has been little reason for utilities to share anything but a small slice of data about their operations with anyone else other than themselves.

Heterogeneous Nuclear Ribonucleoprotein L Negatively Regulates Foot-and-Mouth Disease Virus Replication through Inhibition of Viral RNA Synthesis by Interacting with the Internal Ribosome Entry Site in the 5' Untranslated Region [Virus-Cell Interactio

Upon infection, the highly structured 5' untranslated region (5' UTR) of picornavirus is involved in viral protein translation and RNA synthesis. As a critical element in the 5' UTR, the internal ribosome entry site (IRES) binds to various cellular proteins to function in the processes of picornavirus replication. Foot-and-mouth disease virus (FMDV) is an important member in the family Picornaviridae, and its 5' UTR contains a functional IRES element. In this study, the cellular heterogeneous nuclear ribonucleoprotein L (hnRNP L) was identified as an IRES-binding protein for FMDV by biotinylated RNA pulldown assays, mass spectrometry (MS) analysis, and determination of hnRNP L-IRES interaction regions. Further, we found that hnRNP L inhibited the growth of FMDV through binding to the viral IRES and that the inhibitory effect of hnRNP L on FMDV growth was not due to FMDV IRES-mediated translation, but to influence on viral RNA synthesis. Finally, hnRNP L was demonstrated to coimmunoprecipitate with RNA-dependent RNA polymerase (3Dpol) in an FMDV RNA-dependent manner in the infected cells. Thus, our results suggest that hnRNP L, as a critical IRES-binding protein, negatively regulates FMDV replication by inhibiting viral RNA synthesis, possibly by remaining in the replication complex.

IMPORTANCE Picornaviruses, as a large family of human and animal pathogens, cause a bewildering array of disease syndromes. Many host factors are implicated in the pathogenesis of these viruses, and some proteins interact with the viral IRES elements to affect function. Here, we report for the first time that cellular hnRNP L specifically interacts with the IRES of the picornavirus FMDV and negatively regulates FMDV replication through inhibiting viral RNA synthesis. Further, our results showed that hnRNP L coimmunoprecipitates with FMDV 3Dpol in a viral RNA-dependent manner, suggesting that it may remain in the replication complex to function. The data presented here would facilitate further understanding of virus-host interactions and the pathogenesis of picornavirus infections.

Cosmetic surgery conundrum: is it OK to speculate about Jared Kushner and Botox?

The ‘haunted doll’ look of Donald Trump’s son-in-law has attracted a lot of attention. When people comment on famous women and surgery there is often a backlash, but should the same apply here?

What has happened to Jared Kushner’s face?
Richard, by email

People get a little antsy about the subject of cosmetic surgery: they don’t like to be asked if they have had it, and public speculation over whether someone else has had it is generally considered to be de trop. I don’t really get this. Maybe it’s because I am 100% the world’s worst liar, but pretending to not see that someone’s face has completely changed is a form of magical thinking that is just beyond my capabilities.

Trump finalizes plans to open Utah monuments for mining and drilling

Lawsuits are pending from groups who have challenged the constitutionality of shrinking Bears Ears and Grand Staircase-Escalante

Plans finalized on Thursday for two national monuments in Utah downsized by Donald Trump would ensure that lands previously off-limits to energy development will be open to mining and drilling.

The move comes despite pending lawsuits from conservation, tribal and paleontology groups, who have challenged the constitutionality of the president’s action. The Trump administration slashed the size of Bears Ears national monument by 85% and Grand Staircase-Escalante national monument by nearly half in December 2017, in what represented the largest elimination of public lands protections in US history.

Conservation groups criticized the Trump administration on Thursday for spending time on management plans they believe will become moot when the court sides with their assertion that Trump misused the Antiquities Act to reverse decisions by previous presidents.

RiteAid BonusCash rewards for Apr 5-11, 2020 ... 20% ROI on Xbox, GameStop, Apple, Google, Netflix, Nike, Panera, Fandango, AMC, & Regal GC's

It's a bumper crop of BonusCash at your local Rite-Aid this week, with not 1, 2, 3, but 4 gaming GC's, and 1 of those gives you even more options!

  • Nike, GameStop, Netflix ... $5 BonusCash when you buy $25 of these items.*
  • Google Play, AMC Theatres, Apple AppStore/iTunes, Fandango, XBOX, Panera Bread, Regal Theatres ... $6 BonusCash when you buy $30 of these items.*

FYI, "GameStop" is a big win, because not only can you purchase (additional) XBOX, PSN, Nintendo, and Steam credit there, but you order the GC credit from their website, and get a redemption code instantly after checkout.

For those who are new to the "Rite-Aid wellness+ reward BonusCash" program, you'll receive the $$$ amount when you purchase the minimum amount specified. Gift-cards within the same bullet-point share the same "limit 2 offers per customer", but you can earn rewards on the other bullet-point lines as well. For example, you can purchase $25 each of GameStop & Netflix (or $50 of GameStop) ... and still be able to purchase another $60 mix of Google & Apple & XBOX, and can stagger your 4 GC purchases throughout the week.

Screenshot of 2 separate GC offers (bullet points) included here:

Spoiler

Small print (at bottom of weekly ad) and BonusCash T&C's included here:
Spoiler

FYI ... the limit of "2 offers per customer" is tracked by your "wellness+ rewards" account, so you'll need to limit yourself to 2 offers per line item throughout the week, and not just "2 per transaction" or "2 per day". At the time of purchase, your printed receipt will indicate how many of the "limit 2" you've met, but neither the website nor register will indicate ...

  • if you've met the limit of 2 items per BonusCash group with the current transaction, or
  • if the transaction you're about to complete exceeds the limit of 2 per week, or
  • when your BonusCash rewards will expire.

Luckily the mobile RiteAid app (and website) list your individual accumulation & cashing out on a per transaction basis, so that's a good way to keep tabs on the expiration dates, since you only get 30 days to spend the BonusCash once earned. Good luck!
