Actor, author, and mother Twinkle Khanna on Saturday gave a shout out to all the 'bad-ass mothers' and shared what mother actually wants for the Mother's Day. In a video message which she posted on Twitter, Mrs Funnybones is seen talking about what mothers actually want from their children instead of all the fancy mother's day greetings.

"I am going to tell you what mother's really want for Mother's Day or what at least I want for Mother's Day. I want to be free of all responsibilities for an entire day. I don't want anyone to ask me any questions," says Khanna in the video message.

"Don't ask me where is your blue T-shirt, don't ask me what is 15+73, don't ask me what is going to happen to your 'A' level exams, don't ask me what's for lunch, don't ask me when you can go and meet your friend, don't ask me when the lockdown will lift," she added.

The actor turned author then spoke of how wishing to stay free of any responsibilities makes her feel that she is a 'bad mom' but deep down she also feels that she is a 'bad-ass' mom.

"Some of you watching this may think I am a terrible mom and sometimes I also wonder the same thing especially when my little one looks at me and calls me "Bad Mumma." But deep down I don't think I am a bad mom," the 46-year-old actor said.

"I think that I am a badass mom though I have a perfectly fine posterior. So here's wishing all you badass moms a happy mother's day and the ones with the good ass a happy mother's day to you as well," she added. Twinkle Khanna is a mother to two children -- a son and a daughter.

Twinkle helicase is an essential component of mtDNA replication. Here, we employed atomic force microscopy imaging in air and liquids to visualize ring assembly, DNA binding, and unwinding activity of individual Twinkle hexamers at the single-molecule level. We observed that the Twinkle subunits self-assemble into hexamers and higher-order complexes that can switch between open and closed-ring configurations in the absence of DNA. Our analyses helped visualize Twinkle loading onto and unloading from DNA in an open-ringed configuration. They also revealed that closed-ring conformers bind and unwind several hundred base pairs of duplex DNA at an average rate of ∼240 bp/min. We found that the addition of mitochondrial single-stranded (ss) DNA–binding protein both influences the ways Twinkle loads onto defined DNA substrates and stabilizes the unwound ssDNA product, resulting in a ∼5-fold stimulation of the apparent DNA-unwinding rate. Mitochondrial ssDNA-binding protein also increased the estimated translocation processivity from 1750 to >9000 bp before helicase disassociation, suggesting that more than half of the mitochondrial genome could be unwound by Twinkle during a single DNA-binding event. The strategies used in this work provide a new platform to examine Twinkle disease variants and the core mtDNA replication machinery. They also offer an enhanced framework to investigate molecular mechanisms underlying deletion and depletion of the mitochondrial genome as observed in mitochondrial diseases.

"What mothers really want for mother's day but can't tell their kids," wrote Twinkle

Twinkle Khanna, Hema Malini, Bhumi Pednekar, Kim Sharma, Surbhi Jyoti and many other Bollywood celebrities took it upon themselves to grow their own vegetables. Many have taken to home gardening and decided to grow some veggies and other plants during the lockdown.

The coronavirus outbreak has put many lives and businesses on halt, but people have decided not to give up and find a different way to help each other. Twinkle Khanna grew green veggies in her garden, and sharing the picture, she wrote, "What you sow, you shall reap and I planted a love for gardening in the hearts of my children. This is my harvest-their help in digging, watering and this large bowl of Ceylon Palak. It grows like a weed and is easily propagated by planting the stem back after taking out the leaves. Thank you @merliynjoseph for helping me with this a few years ago. It is a gift that keeps giving in so many ways. #greenthumb [sic]"

Flaunting her gardening skills, actor and writer Twinkle Khanna on Wednesday treated all the nature lovers with a picture of her perfectly organised and well-groomed garden.

The actor with the 'green thumb' took to Instagram to post the picture and also described how she is clumsy at all other chores like cooking, but has spectacular gardening skills.

The picture featured a nicely kept garden with greenery spilling from every corner.

The central attraction of the green nest was the sitting area that had a wooden table with a big plant on it and chairs kept around the table.

The garden further had fresh flowers fallen from the trees all around on the ground adding to the perfect natural aesthetics.
