gp130 functions as a shared signal-transducing subunit not only for interleukin (IL)-6 but also for eight other human cytokine receptor complexes. The IL-6 signaling pathway mediated through gp130 encompasses classical, trans, or cluster signaling, intricately regulated by a diverse array of modulators affecting IL-6, its receptor, and gp130. Currently, only a limited number of small molecule antagonists and agonists for gp130 are known. This review aims to comprehensively examine the current knowledge of these modulators and provide insights into their pharmacological properties, particularly in the context of cancer and other diseases. Notably, the prominent gp130 modulators SC144, bazedoxifene, and raloxifene are discussed in detail, with a specific focus on the discovery of SC144’s iron-chelating properties. This adds a new dimension to the understanding of its pharmacological effects and therapeutic potential in conditions where iron homeostasis is significant. Our bioinformatic analysis of gp130 and genes related to iron homeostasis reveals insightful correlations, implicating the role of iron in the gp130 signaling pathway. Overall, this review contributes to the evolving understanding of gp130 modulation and its potential therapeutic applications in various disease contexts.

A recent overview and analysis shows that increasing amounts of gas drilling at Groningen, the largest gas field in Europe, led to a dramatic rise in regional earthquakes between 2001 and 2013. After a reduction in extraction was introduced by the Dutch Government, earthquake numbers started to fall. Statistical analysis reveals that if high extraction rates were resumed, about 35 earthquakes, with a magnitude (M) of over 1.5 on the Richter scale, might occur annually from the year 2021 onwards, including four with a damaging magnitude of over 2.5.

A component composition contains, by % by mass, 0.0016 to 0.01% of C, 0.05 to 0.60% of Mn, and 0.020 to 0.080% of Nb so that the C and Nb contents satisfy the expression, 0.4≦(Nb/C)×(12/93)≦2.5. This invention relates to the ironing of fabrics, such as clothes in particular, and provides a compact, light-weight apparatus suitable also for use in various ironing applications. The ironing apparatus comprises first and second fabric-pressing surfaces, supporting arms to which the surfaces are mounted opposite each other, to define a fabric-receiving gap between the surfaces. The supporting arms operate to urge the first surface toward the second, so to narrow the gap from a first, fabric receiving width, to a second, fabric-compressing width effective for exerting crease-reducing pressure on said fabric in the gap, while permitting relative traversing movement between said surfaces and the fabric. The supporting arms may be in the form of a flexible U-shaped member having opposed ends to which the respective pressing surfaces are mounted to be inwardly opposed, and a generally central portion defining a body.

Mutation and translocation of fibroblast growth factor receptors often lead to aberrant signaling and cancer. This work focuses on the t(8;22)(p11;q11) chromosomal translocation which creates the breakpoint cluster region (BCR) fibroblast growth factor receptor1 (FGFR1) (BCR-FGFR1) fusion protein. This fusion occurs in stem cell leukemia/lymphoma, which can progress to atypical chronic myeloid leukemia, acute myeloid leukemia, or B-cell lymphoma. This work focuses on the biochemical characterization of BCR-FGFR1 and identification of novel therapeutic targets. The tyrosine kinase activity of FGFR1 is required for biological activity as shown using transformation assays, interleukin-3 independent cell proliferation, and liquid chromatography/mass spectroscopy analyses. Furthermore, BCR contributes a coiled-coil oligomerization domain, also essential for oncogenic transformation by BCR-FGFR1. The importance of salt bridge formation within the coiled-coil domain is demonstrated, as disruption of three salt bridges abrogates cellular transforming ability. Lastly, BCR-FGFR1 acts as a client of the chaperonin heat shock protein 90 (Hsp90), suggesting that BCR-FGFR1 relies on Hsp90 complex to evade proteasomal degradation. Transformed cells expressing BCR-FGFR1 are sensitive to the Hsp90 inhibitor Ganetespib, and also respond to combined treatment with Ganetespib plus the FGFR inhibitor BGJ398. Collectively, these data suggest novel therapeutic approaches for future stem cell leukemia/lymphoma treatment: inhibition of BCR oligomerization by disruption of required salt bridges; and inhibition of the chaperonin Hsp90 complex.
