Trailer for EON's New Spy Movie THE RHYTHM SECTION

On Friday Paramount dropped the trailer for the second most anticipated EON Production of 2020, The Rhythm Section! The Rhythm Section has been delayed several times (first when star Blake Lively suffered an on-set injury), but here's proof that it's finally really coming... and it looks great! While an adaptation of Mark Burnell's 1999 spy novel would be something for spy fans to be seriously excited about anyway, it's even more exciting because it hails from Barbara Broccoli and Michael G. Wilson's EON Productions, the producers behind the James Bond movies. While EON has been venturing outside the realm of 007 lately, this marks their first new foray into the genre that defined them—and that they defined, under the auspices of first-generation Bond producers Albert R. Broccoli and Harry Saltzman. And it's not only a new EON spy movie; it's potentially the start of a new, female-fronted EON spy series! (Burnell wrote four Stephanie Patrick thrillers.) Will Lively end up being the Sean Connery of a long lasting Stephanie Patrick film series?

The books are quite good, and remind me of a female Callan. Like Callan, Stephanie ends up working as an assassin for a particularly unpleasant boss in an ultra-secret branch of British Intelligence. And like Callan, she doesn't do this work by choice. Instead she's forced into it by that unpleasant boss. But she's also got very personal motivations (motivations he ruthlessly manipulates) for her initial mission: an opportunity to get revenge on the terrorists responsible for the death of her parents and siblings. Burnell's book is very dark and very serious, and judging from this trailer the movie will be true to that tone. In fact, the movie (directed by Reed Morano and scripted by Burnell himself) looks quite faithful to the book overall, though it's obvious that the ending has been changed, which was pretty much a given. (The villains' plot in the '99 book had eerie similarities to 9/11, which simply wouldn't play in today's world.) And it looks great!

The first of two major EON spy movies coming out next year, The Rhythm Section opens on January 31, 2020. It stars Blake Lively (The Age of Adaline), Jude Law (Spy), Raza Jaffrey (Spooks/MI-5), and Sterling K. Brown (Black Panther).

The Arg-293 of Cryptochrome1 is responsible for the allosteric regulation of CLOCK-CRY1 binding in circadian rhythm [Computational Biology]

Mammalian circadian clocks are driven by transcription/translation feedback loops composed of positive transcriptional activators (BMAL1 and CLOCK) and negative repressors (CRYPTOCHROMEs (CRYs) and PERIODs (PERs)). CRYs, in complex with PERs, bind to the BMAL1/CLOCK complex and repress E-box–driven transcription of clock-associated genes. There are two individual CRYs, with CRY1 exhibiting higher affinity to the BMAL1/CLOCK complex than CRY2. It is known that this differential binding is regulated by a dynamic serine-rich loop adjacent to the secondary pocket of both CRYs, but the underlying features controlling loop dynamics are not known. Here we report that allosteric regulation of the serine-rich loop is mediated by Arg-293 of CRY1, identified as a rare CRY1 SNP in the Ensembl and 1000 Genomes databases. The p.Arg293His CRY1 variant caused a shortened circadian period in a Cry1−/−Cry2−/− double knockout mouse embryonic fibroblast cell line. Moreover, the variant displayed reduced repressor activity on BMAL1/CLOCK driven transcription, which is explained by reduced affinity to BMAL1/CLOCK in the absence of PER2 compared with CRY1. Molecular dynamics simulations revealed that the p.Arg293His CRY1 variant altered a communication pathway between Arg-293 and the serine loop by reducing its dynamicity. Collectively, this study provides direct evidence that allosterism in CRY1 is critical for the regulation of circadian rhythm.

Analysis of Sleep and Circadian Rhythms from Drosophila Activity-Monitoring Data Using SCAMP

Sleep is a fundamental feature of life for virtually all multicellular animals, but many questions remain about how sleep is regulated and what biological functions it plays. Substantial headway has been made in the study of both circadian rhythms and sleep in the fruit fly Drosophila melanogaster, much of it through studies of individual fly activity using beam break counts from Drosophila activity monitors (DAMs). The number of laboratories worldwide studying sleep in Drosophila has grown from only a few 20 years ago to hundreds today. The utility of these studies is limited by the quality of the metrics that can be extracted from the data. Many software options exist to help analyze DAM data; however, these are often expensive or have significant limitations. Therefore, we describe here a method for analyzing DAM-based data using the sleep and circadian analysis MATLAB program (SCAMP). This user-friendly software has an advantage of combining several analyses of both sleep and circadian rhythms in one package and produces graphical outputs as well as spreadsheets of the outputs for further statistical analysis. The version of SCAMP described here is also the first published software package that can analyze data from multibeam DAM5Ms, enabling determination of positional preference over time.

Neural Stimulation during Drosophila Activity Monitor (DAM)-Based Studies of Sleep and Circadian Rhythms in Drosophila melanogaster

Sleep is a fundamental feature of life for virtually all multicellular animals, but many questions remain about how sleep is regulated by circadian rhythms, homeostatic sleep drive that builds up with wakefulness, and modifying factors such as hunger or social interactions, as well as about the biological functions of sleep. Substantial headway has been made in the study of both circadian rhythms and sleep in the fruit fly Drosophila melanogaster, much of it through studies of individual fly activity using Drosophila activity monitors (DAMs). Here, we describe approaches for the activation of specific neurons of interest using optogenetics (involving genetic modifications that allow for light-based neuronal activation) and thermogenetics (involving genetic modifications that allow for temperature-based neuronal activation) so that researchers can evaluate the roles of those neurons in controlling rest and activity behavior. In this protocol, we describe how to set up a rig for simultaneous optogenetic or thermogenetic stimulation and activity monitoring for analysis of sleep and circadian rhythms in Drosophila, how to raise appropriate flies, and how to perform the experiment. This protocol will allow researchers to assess the causative role in the regulation of sleep and activity rhythms of any genetically tractable subset of cells.

Event-related desynchronization and synchronization (ERD/S) and movement-related cortical potential (MRCP) play an important role in brain-computer interfaces (BCI) for lower limb rehabilitation, particularly in standing and sitting. However, little is known about the differences in the cortical activation between standing and sitting, especially how the brain's intention modulates the pre-movement sensorimotor rhythm as they do for switching movements. In this study, we aim to investigate the decoding of continuous EEG rhythms during action observation (AO), motor imagery (MI), and motor execution (ME) for standing and sitting. We developed a behavioral task in which participants were instructed to perform both AO and MI/ME in regard to the actions of sit-to-stand and stand-to-sit. Our results demonstrated that the ERD was prominent during AO, whereas ERS was typical during MI at the alpha band across the sensorimotor area. A combination of the filter bank common spatial pattern (FBCSP) and support vector machine (SVM) for classification was used for both offline and pseudo-online analyses. The offline analysis indicated the classification of AO and MI providing the highest mean accuracy at 82.73$pm$2.38\% in stand-to-sit transition. By applying the pseudo-online analysis, we demonstrated the higher performance of decoding neural intentions from the MI paradigm in comparison to the ME paradigm. These observations led us to the promising aspect of using our developed tasks based on the integration of both AO and MI to build future exoskeleton-based rehabilitation systems.

Apparatus for detecting and treating ventricular arrhythmia

A system and method for long-term monitoring of cardiac conditions such as arrhythmias is disclosed. The invention includes a pulse generator including means for sensing an arrhythmia. The pulse generator is coupled to at least one subcutaneous electrode or electrode array for providing electrical stimulation such as cardioversion/defibrillation shocks and/or pacing pulses. The electrical stimulation may be provided between multiple subcutaneous electrodes, or between one or more such electrodes and the housing of the pulse generator. In one embodiment, the pulse generator includes one or more electrodes that are isolated from the can. These electrodes may be used to sense cardiac signals.
