Ironing Out the Mechanism of gp130 Signaling [Review Article]

gp130 functions as a shared signal-transducing subunit not only for interleukin (IL)-6 but also for eight other human cytokine receptor complexes. The IL-6 signaling pathway mediated through gp130 encompasses classical, trans, or cluster signaling, intricately regulated by a diverse array of modulators affecting IL-6, its receptor, and gp130. Currently, only a limited number of small molecule antagonists and agonists for gp130 are known. This review aims to comprehensively examine the current knowledge of these modulators and provide insights into their pharmacological properties, particularly in the context of cancer and other diseases. Notably, the prominent gp130 modulators SC144, bazedoxifene, and raloxifene are discussed in detail, with a specific focus on the discovery of SC144’s iron-chelating properties. This adds a new dimension to the understanding of its pharmacological effects and therapeutic potential in conditions where iron homeostasis is significant. Our bioinformatic analysis of gp130 and genes related to iron homeostasis reveals insightful correlations, implicating the role of iron in the gp130 signaling pathway. Overall, this review contributes to the evolving understanding of gp130 modulation and its potential therapeutic applications in various disease contexts.

Intestinal Lymphatic Biology, Drug Delivery, and Therapeutics: Current Status and Future Directions [Review Article]

Historically, the intestinal lymphatics were considered passive conduits for fluids, immune cells, dietary lipids, lipid soluble vitamins, and lipophilic drugs. Studies of intestinal lymphatic drug delivery in the late 20th century focused primarily on the drugs’ physicochemical properties, especially high lipophilicity, that resulted in intestinal lymphatic transport. More recent discoveries have changed our traditional view by demonstrating that the lymphatics are active, plastic, and tissue-specific players in a range of biological and pathological processes, including within the intestine. These findings have, in turn, inspired exploration of lymph-specific therapies for a range of diseases, as well as the development of more sophisticated strategies to actively deliver drugs or vaccines to the intestinal lymph, including a range of nanotechnologies, lipid prodrugs, and lipid-conjugated materials that "hitchhike" onto lymphatic transport pathways. With the increasing development of novel therapeutics such as biologics, there has been interest in whether these therapeutics are absorbed and transported through intestinal lymph after oral administration. Here we review the current state of understanding of the anatomy and physiology of the gastrointestinal lymphatic system in health and disease, with a focus on aspects relevant to drug delivery. We summarize the current state-of-the-art approaches to deliver drugs and quantify their uptake into the intestinal lymphatic system. Finally, and excitingly, we discuss recent examples of significant pharmacokinetic and therapeutic benefits achieved via intestinal lymphatic drug delivery. We also propose approaches to advance the development and clinical application of intestinal lymphatic delivery strategies in the future.

Somatostatin: Linking Cognition and Alzheimer Disease to Therapeutic Targeting [Review Article]

Over 4 decades of research support the link between Alzheimer disease (AD) and somatostatin [somatotropin-releasing inhibitory factor (SRIF)]. SRIF and SRIF-expressing neurons play an essential role in brain function, modulating hippocampal activity and memory formation. Loss of SRIF and SRIF-expressing neurons in the brain rests at the center of a series of interdependent pathological events driven by amyloid-β peptide (Aβ), culminating in cognitive decline and dementia. The connection between the SRIF and AD further extends to the neuropsychiatric symptoms, seizure activity, and inflammation, whereas preclinical AD investigations show SRIF or SRIF receptor agonist administration capable of enhancing cognition. SRIF receptor subtype-4 activation in particular presents unique attributes, with the potential to mitigate learning and memory decline, reduce comorbid symptoms, and enhance enzymatic degradation of Aβ in the brain. Here, we review the links between SRIF and AD along with the therapeutic implications.

Significance Statement

Somatostatin and somatostatin-expressing neurons in the brain are extensively involved in cognition. Loss of somatostatin and somatostatin-expressing neurons in Alzheimer disease rests at the center of a series of interdependent pathological events contributing to cognitive decline and dementia. Posttranslational Modifications of {alpha}-Synuclein, Their Therapeutic Potential, and Crosstalk in Health and Neurodegenerative Diseases [Review Article]

α-Synuclein (α-Syn) aggregation in Lewy bodies and Lewy neurites has emerged as a key pathogenetic feature in Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. Various factors, including posttranslational modifications (PTMs), can influence the propensity of α-Syn to misfold and aggregate. PTMs are biochemical modifications of a protein that occur during or after translation and are typically mediated by enzymes. PTMs modulate several characteristics of proteins including their structure, activity, localization, and stability. α-Syn undergoes various posttranslational modifications, including phosphorylation, ubiquitination, SUMOylation, acetylation, glycation, O-GlcNAcylation, nitration, oxidation, polyamination, arginylation, and truncation. Different PTMs of a protein can physically interact with one another or work together to influence a particular physiological or pathological feature in a process known as PTMs crosstalk. The development of detection techniques for the cooccurrence of PTMs in recent years has uncovered previously unappreciated mechanisms of their crosstalk. This has led to the emergence of evidence supporting an association between α-Syn PTMs crosstalk and synucleinopathies. In this review, we provide a comprehensive evaluation of α-Syn PTMs, their impact on misfolding and pathogenicity, the pharmacological means of targeting them, and their potential as biomarkers of disease. We also highlight the importance of the crosstalk between these PTMs in α-Syn function and aggregation. Insight into these PTMS and the complexities of their crosstalk can improve our understanding of the pathogenesis of synucleinopathies and identify novel targets of therapeutic potential.

Significance Statement

α-Synuclein is a key pathogenic protein in Parkinson’s disease and other synucleinopathies, making it a leading therapeutic target for disease modification. Multiple posttranslational modifications occur at various sites in α-Synuclein and alter its biophysical and pathological properties, some interacting with one another to add to the complexity of the pathogenicity of this protein. This review details these modifications, their implications in disease, and potential therapeutic opportunities.

Bile Acid Signaling in Metabolic and Inflammatory Diseases and Drug Development [Review Article]

Bile acids are the end products of cholesterol catabolism. Hepatic bile acid synthesis accounts for a major fraction of daily cholesterol turnover in humans. Biliary secretion of bile acids generates bile flow and facilitates biliary secretion of lipids, endogenous metabolites, and xenobiotics. In intestine, bile acids facilitate the digestion and absorption of dietary lipids and fat-soluble vitamins. Through activation of nuclear receptors and G protein-coupled receptors and interaction with gut microbiome, bile acids critically regulate host metabolism and innate and adaptive immunity and are involved in the pathogenesis of cholestasis, metabolic dysfunction-associated steatotic liver disease, alcohol-associated liver disease, type-2 diabetes, and inflammatory bowel diseases. Bile acids and their derivatives have been developed as potential therapeutic agents for treating chronic metabolic and inflammatory liver diseases and gastrointestinal disorders.

Significance Statement

Bile acids facilitate biliary cholesterol solubilization and dietary lipid absorption, regulate host metabolism and immunity, and modulate gut microbiome. Targeting bile acid metabolism and signaling holds promise for treating metabolic and inflammatory diseases.

Glatiramer Acetate for the Treatment of Multiple Sclerosis: From First-Generation Therapy to Elucidation of Immunomodulation and Repair [Review Article]

Multiple sclerosis (MS) is a chronic inflammatory demyelinating and neurodegenerative disease of the central nervous system (CNS), with a putative autoimmune origin and complex pathogenesis. Modification of the natural history of MS by reducing relapses and slowing disability accumulation was first attained in the 1990 s with the development of the first-generation disease-modifying therapies. Glatiramer acetate (GA), a copolymer of L-alanine, L-lysine, L-glutamic acid, and L-tyrosine, was discovered due to its ability to suppress the animal model of MS, experimental autoimmune encephalomyelitis. Extensive clinical trials and long-term assessments established the efficacy and the safety of GA. Furthermore, studies of the therapeutic processes induced by GA in animal models and in MS patients indicate that GA affects various levels of the innate and the adaptive immune response, generating deviation from proinflammatory to anti-inflammatory pathways. This includes competition for binding to antigen presenting cells; driving dendritic cells, monocytes, and B-cells toward anti-inflammatory responses; and stimulating T-helper 2 and T-regulatory cells. The immune cells stimulated by GA reach the CNS and secrete in situ anti-inflammatory cytokines alleviating the pathological processes. Furthermore, cumulative findings reveal that in addition to its immunomodulatory effect, GA promotes neuroprotective repair processes such as neurotrophic factors secretion, remyelination, and neurogenesis. This review aims to provide an overview of MS pathology diagnosis and treatment as well as the diverse mechanism of action of GA.

Significance Statement

Understanding the complex MS immune pathogenesis provided multiple targets for therapeutic intervention, resulting in a plethora of agents, with various mechanisms of action, efficacy, and safety profiles. However, promoting repair beyond the body’s limited spontaneous extent is still a major challenge. GA, one of the first approved disease-modifying therapies, induces diverse immunomodulatory effects. Furthermore, GA treatment results in elevated neurotrophic factors secretion, remyelination and neurogenesis, supporting the notion that immunomodulatory treatment can support in situ a growth-promoting and repair environment.

Roles of Individual Human Cytochrome P450 Enzymes in Drug Metabolism [Review Article]

Our knowledge of the roles of individual cytochrome P450 (P450) enzymes in drug metabolism has developed considerably in the past 30 years, and this base has been of considerable use in avoiding serious issues with drug interactions and issues due to variations. Some newer approaches are being considered for "phenotyping" metabolism reactions with new drug candidates. Endogenous biomarkers are being used for noninvasive estimation of levels of individual P450 enzymes. There is also the matter of some remaining "orphan" P450s, which have yet to be assigned reactions. Practical problems that continue in drug development include predicting drug-drug interactions, predicting the effects of polymorphic and other P450 variations, and evaluating interspecies differences in drug metabolism, particularly in the context of "metabolism in safety testing" regulatory issues ["disproportionate (human) metabolites"].

Significance Statement

Cytochrome P450 enzymes are the major catalysts involved in drug metabolism. The characterization of their individual roles has major implications in drug development and clinical practice.

The 75-Year Anniversary of the Department of Physiology and Pharmacology at Karolinska Institutet--Examples of Recent Accomplishments and Future Perspectives [75th Anniversary Celebration Collection Special Section-Perspective]

Karolinska Institutet is a medical university encompassing 21 departments distributed across three departmental or campus groups. Pharmacological research has a long and successful tradition at the institute with a multitude of seminal findings in the areas of neuronal control of vasodilatation, cardiovascular pharmacology, neuropsychopharmacology, receptor pharmacology, and pharmacogenomics that resulted in, among many other recognitions, two Nobel prizes in Physiology and Medicine, one in 1970 to Ulf von Euler for his discovery of the processes involved in storage, release, and inactivation of neurotransmitters and the other in 1982 to Sune Bergström and Bengt Samuelsson for their work on prostaglandins and the discovery of leukotrienes. Pharmacology at Karolinska Institutet has over the last decade been ranked globally among the top 10 according to the QS World University Ranking. With the Department of Physiology and Pharmacology now celebrating its 75-year anniversary, we wanted to take this as an opportunity to showcase recent research achievements and how they paved the way for current activities at the department. We emphasize examples from preclinical and clinical research where the dpartment's integrative environment and robust infrastructure have successfully facilitated the translation of findings into clinical applications and patient benefits. The close collaboration between preclinical scientists and clinical researchers across various disciplines, along with a strong network of partnerships within the department and beyond, positions us to continue leading world-class pharmacological research at the Department of Physiology and Pharmacology for decades to come.

Significance Statement

Pharmacological research at Karolinska Institutet has a long and successful history. Given the 75-year anniversary of the Department of Physiology and Pharmacology, this perspective provides an overview of recent departmental achievements and future trajectories. Pharmacological Approaches to Hearing Loss [75th Anniversary Celebration Collection Special Section]

Hearing disorders pose significant challenges to individuals experiencing them and their overall quality of life, emphasizing the critical need for advanced pharmacological approaches to address these conditions. Current treatment options often focus on amplification devices, cochlear implants, or other rehabilitative therapies, leaving a substantial gap regarding effective pharmacological interventions. Advancements in our understanding of the molecular and cellular mechanisms involved in hearing disorders induced by noise, aging, and ototoxicity have opened new avenues for drug development, some of which have led to numerous clinical trials, with promising results. The development of optimal drug delivery solutions in animals and humans can also enhance the targeted delivery of medications to the ear. Moreover, large genome studies contributing to a genetic understanding of hearing loss in humans combined with advanced molecular technologies in animal studies have shown a great potential to increase our understanding of the etiologies of hearing loss. The auditory system exhibits circadian rhythms and temporal variations in its physiology, its vulnerability to auditory insults, and its responsiveness to drug treatments. The cochlear clock rhythms are under the control of the glucocorticoid system, and preclinical evidence suggests that the risk/benefit profile of hearing disorder treatments using chronopharmacological approaches would be beneficial. If translatable to the bedside, such approaches may improve the outcome of clinical trials. Ongoing research into the molecular and genetic basis of auditory disorders, coupled with advancements in drug formulation and delivery as well as optimized timing of drug administration, holds great promise of more effective treatments.

Nitric Oxide Signaling and Regulation in the Cardiovascular System: Recent Advances [75th Anniversary Celebration Collection Special Section]

Nitric oxide (NO) from endothelial NO synthase importantly contributes to vascular homeostasis. Reduced NO production or increased scavenging during disease conditions with oxidative stress contribute to endothelial dysfunction and NO deficiency. In addition to the classical enzymatic NO synthases (NOS) system, NO can also be generated via the nitrate-nitrite-NO pathway. Dietary and pharmacological approaches aimed at increasing NO bioactivity, especially in the cardiovascular system, have been the focus of much research since the discovery of this small gaseous signaling molecule. Despite wide appreciation of the biological role of NOS/NO signaling, questions still remain about the chemical nature of NOS-derived bioactivity. Recent studies show that NO-like bioactivity can be efficiently transduced by mobile NO-ferroheme species, which can transfer between proteins, partition into a hydrophobic phase, and directly activate the soluble guanylyl cyclase-cGMP-protein kinase G pathway without intermediacy of free NO. Moreover, interaction between red blood cells and the endothelium in the regulation of vascular NO homeostasis have gained much attention, especially in conditions with cardiometabolic disease. In this review we discuss both classical and nonclassical pathways for NO generation in the cardiovascular system and how these can be modulated for therapeutic purposes.

Significance Statement

After four decades of intensive research, questions persist about the transduction and control of nitric oxide (NO) synthase bioactivity. Here we discuss NO signaling in cardiovascular health and disease, highlighting new findings, such as the important role of red blood cells in cardiovascular NO homeostasis. Nonclassical signaling modes, like the nitrate-nitrite-NO pathway, and therapeutic opportunities related to the NO system are discussed. Existing and potential pharmacological treatments/strategies, as well as dietary components influencing NO generation and signaling are covered.

International Union of Basic and Clinical Pharmacology CXV: The Class F of G Protein-Coupled Receptors [75th Anniversary Celebration Collection Special Section]

The class F of G protein-coupled receptors (GPCRs) consists of 10 Frizzleds (FZD1–10) and Smoothened (SMO). FZDs bind and are activated by secreted lipoglycoproteins of the Wingless/Int-1 (WNT) family, and SMO is indirectly activated by the Hedgehog (Hh) family of morphogens acting on the transmembrane protein Patched. The advance of our understanding of FZDs and SMO as dynamic transmembrane receptors and molecular machines, which emerged during the past 14 years since the first-class F GPCR IUPHAR nomenclature report, justifies an update. This article focuses on the advances in molecular pharmacology and structural biology providing new mechanistic insight into ligand recognition, receptor activation mechanisms, signal initiation, and signal specification. Furthermore, class F GPCRs continue to develop as drug targets, and novel technologies and tools such as genetically encoded biosensors and CRISP/Cas9 edited cell systems have contributed to refined functional analysis of these receptors. Also, advances in crystal structure analysis and cryogenic electron microscopy contribute to the rapid development of our knowledge about structure-function relationships, providing a great starting point for drug development. Despite the progress, questions and challenges remain to fully understand the complexity of the WNT/FZD and Hh/SMO signaling systems.

Significance Statement

The recent years of research have brought about substantial functional and structural insight into mechanisms of activation of Frizzleds and Smoothened. While the advance furthers our mechanistic understanding of ligand recognition, receptor activation, signal specification, and initiation, broader opportunities emerge that allow targeting class F GPCRs for therapy and regenerative medicine employing both biologics and small molecule compounds.

Neuroactive Kynurenines as Pharmacological Targets: New Experimental Tools and Exciting Therapeutic Opportunities [75th Anniversary Celebration Collection Special Section]

Both preclinical and clinical studies implicate functional impairments of several neuroactive metabolites of the kynurenine pathway (KP), the major degradative cascade of the essential amino acid tryptophan in mammals, in the pathophysiology of neurologic and psychiatric diseases. A number of KP enzymes, such as tryptophan 2,3-dioxygenase (TDO2), indoleamine 2,3-dioxygenases (IDO1 and IDO2), kynurenine aminotransferases (KATs), kynurenine 3-monooxygenase (KMO), 3-hydroxyanthranilic acid oxygenase (3-HAO), and quinolinic acid phosphoribosyltransferase (QPRT), control brain KP metabolism in health and disease and are therefore increasingly considered to be promising targets for the treatment of disorders of the nervous system. Understanding the distribution, cellular expression, and regulation of KP enzymes and KP metabolites in the brain is therefore critical for the conceptualization and implementation of successful therapeutic strategies.

Significance Statement

Studies have implicated the kynurenine pathway of tryptophan in the pathophysiology of neurologic and psychiatric diseases. Key enzymes of the kynurenine pathway regulate brain metabolism in both health and disease, making them promising targets for treating these disorders. Therefore, understanding the distribution, cellular expression, and regulation of these enzymes and metabolites in the brain is critical for developing effective therapeutic strategies. Jimmy Savile and Newsnight: A correction

The following is a statement issued by the BBC

The BBC has launched an independent review, led by former Head of Sky News Nick Pollard, to determine whether there were any failings in the BBC's management of the Newsnight investigation into allegations of sexual abuse of children by Jimmy Savile.

However, on the basis of material available now, it is apparent from information supplied by the Newsnight editor and programme team - that the explanation in a blog by the editor of his decision to drop the programme's investigation is inaccurate or incomplete in some respects.

By way of correction and clarification:

1.The blog says that Newsnight had no evidence that anyone from the Duncroft home could or should have known about the allegations. In fact some allegations were made (mostly in general terms) that some of the Duncroft staff knew or may have known about the abuse.

2. The blog says that Newsnight had no evidence against the BBC. No allegation was made to the programme that BBC staff were aware of Mr Savile's alleged activities, but there were some allegations of abusive conduct on BBC premises.

3. The blog says that all the women spoken to by the programme had contacted the police independently already and that Newsnight had no new evidence against any other person that would have helped the police. It appears that in some cases women had not spoken to the police and that the police were not aware of all the allegations.

The BBC regrets these errors and will work with the Pollard review to assemble all relevant evidence to enable the review to determine the full facts.

Earlier this year we launched a new version of the BBC News mobile site, making it easier and quicker to use. This week we've begun the process of directing all mobile users automatically to that site. This means that anyone who visits BBC News on their mobile will be taken to the version of the site best suited to the type of phone they are using.

Many of you already visit the mobile site regularly but, up until now, people looking for BBC News on their phone will often have found themselves on the desktop version of the site, which is designed for desktop PCs, macs and laptops - all with much bigger screens. If you are using this desktop version on your phone it can be awkward to pinch, zoom and scan the stories on a small mobile screen.

To tackle this, we've been working over the past six months to improve and add to the mobile site, taking on board your feedback about how you'd like to see it develop.

We've recently added video clips for iPhone and Android users, and made it easier to navigate the site. (We hope to extend this video service to other types of mobile in the future.) We've also added easier ways of getting to local news and weather services, something many of you asked for. You can read more about those changes here.

So, we're confident that the mobile site now has the wide range of content you are looking for and that it offers a better experience on a small screen than the desktop site, which is why we are taking the step of automatically redirecting mobile users there.

Of course, you may be happy to keep visiting the desktop site on your mobile and if you want to continue doing so just scroll to the bottom of the page and tap on the link for the desktop site. Your choice will be remembered for the next time you visit.

Similarly, if you use a mobile and find that you're not redirected to the new site, you can scroll to the bottom of the page and select the mobile site.

This is the latest stage in the ongoing work by our News product team on responsive design - a way of presenting our content to you in the most suitable way by detecting the type of device you are using and displaying the format best adapted for it. We are doing similar work to optimise the site for tablet users too.

The number of people coming to BBC News on mobile continues to grow. In an average week, 13.3m users worldwide use their mobile or tablet to visit the BBC News site and apps - around one-third of total users to BBC News Online.

If you are one of them, our aim is to offer you the full range and depth of BBC News content, whatever device you are using, whilst also making best use of the screen size.

John Walsh – Kindle: As a tablet device, albeit with a smaller screen than some makes, Kindles currently default to the desktop site. Users of any device including Kindles are certainly free to use the mobile version if they prefer by clicking the link at the bottom of the screen. Our aim is to further improve the experience for progressively larger screen sizes over time.

Jesse Moore - HTC: We know there are some devices that are incorrectly classified by our systems, often due to the fact that some devices have different identifiers dependent on the mobile network they are on. In any case we will certainly be doing everything we can to correct errors and ensure the redirection behaves as it should. In the meantime, please use the “Mobile Site” link at the foot of the page should you wish to use the mobile site – the selection will be remembered as long as cookies are not cleared. At this time the redirect only applies to the BBC Homepage and the News site.

Cogito Ergo Sum - Windows phone: This change applies to the browser experience, which is already designed to work for Windows Phone although at present we are unable to provide video for those devices.

Costmeabob - We take accessibility for our services seriously so, for example, our browser and applications are designed to work with Voiceover on iOS. 

Tim Stey - If you do still prefer the desktop version you can select the link at the bottom of the page and you’ll be taken to it. Your choice will be remembered next time you visit the site. Goodbye Ceefax

 

Ceefax - the BBC's teletext service - finally ends its long career tonight when it is due to be switched off at 23:30 BST. There is more on this, and the history of the service, in our news story today and linked coverage.

As each part of the UK has in turn gone through the switchover to digital and lost the Ceefax service in the process, it has been a long farewell, which I have written about here before.

Now, with the analogue TV signal in Northern Ireland being switched off, the last stage in the process has arrived, and the service will come to an end.

The BBC Red Button services will carry on the Ceefax tradition of providing clear and concise news from around the UK and the world, on demand, on your TV.

Indeed the Red Button service is in the process of being reinvented for internet-enabled TV sets, and this “Connected Red Button” service will combine the simplicity of traditional Red Button with the flexibility and depth of online. My colleague Daniel Danker has written about this work here and there is already a BBC News app for connected TVs which I wrote about here and here when it launched.

At its peak, Ceefax had an audience of some 20 million viewers a week, and as the end of the service has approached, it has received several thousand letters and emails of thanks from viewers.

In a tribute to the clarity of Ceefax’s simple, concise format and news stories, and to mark Ceefax's last day, the Plain English Campaign - which campaigns for clear, concise language in public information - has given Ceefax a lifetime achievement award.

It's an honour to have received so many tributes from Ceefax viewers, and to get this award, and both are a recognition of the skill and dedication of all the journalists who have worked on the service over the years, and the care they have taken in writing every story.

Election night

So after more than a year of campaigning, it all comes down to this. On radio, TV and online, the BBC is gearing up for a big night - in English and 27 other languages. And not just one big night, but 51 separate contests.

Unlike most other countries, the US election is not a nationwide "popular poll". Instead, the president is elected by an Electoral College of 538 delegates from each of the 50 states, plus the District of Columbia. How many come from each, depends on their population. So as the votes pile up, it's the way each state votes that will decide the election.

In most states, thanks to exit polls, it may be possible to project a result the moment the polls close. Working with our friends at ABC News, the BBC will "call" the results, state by state, based on those projections. In states that are too close to call, electronic voting will mean we're able to follow the counting in real time, based on the number of voting precincts reporting.

The first real test will come at midnight GMT when polls close in six states. Virginia, with 13 electoral college votes, will be the first of the battle ground states to report. Half an hour later at 00:30 GMT, polls will also close in Ohio with its 18 votes and North Carolina with 15 votes. As the polls close, the BBC will call the result in each based on projections made by ABC News.

Using the results service on the BBC News website, you'll be able to follow the same data driving the BBC's results system on TV and radio. They will include the state results, the resultant change in the Electoral College vote, and will colour the state and national maps accordingly - red for Republican states, blue for Democrats.

The target for Barack Obama and Mitt Romney is to hit a figure of 270 - winning the majority of the 538 delegates to the Electoral College. Once one of the candidates passes the magic 270 total, this election will be over. Then - and only then - will the BBC call the election. A big night and, possibly, a long night beckons.

Election stats - new mobile record

BBC coverage of the US election, which my colleague Jon Williams trailed here a couple of days ago, brought the highest traffic to BBC News Online so far this year, and set a new record for us on mobile.

On 7 November, there were 16.4m unique browsers across the website and mobile, 8.1m of which came from the UK. That makes it the highest traffic day of 2012 so far and rivals our two biggest previous days during the August riots and the March Tsunami, in 2011. During the England riots, on 9 August 2011 there were there were 18.2m unique browsers, 10.9m of which came from the UK.

The peak traffic point yesterday was 07:00-08:00 GMT, which saw higher usage than lunchtime, maybe as people checked the results as soon as they woke up. UK usage figures yesterday were 50% higher than the average for 2012, and ex-UK usage was 75% higher than average.

We spent a lot of time working out how to provide the best possible service on mobile, so it's encouraging to see that nearly 5m mobile devices visited BBC News Online yesterday, a record figure for us on mobile, accounting for about 30% of all users yesterday (on an average weekday, we'd expect mobiles to account for about 24% of users).

Mozilla Festival and the fellowship announcement

Back in July we announced that we'd be working with the Knight-Mozilla fellowship for a second year and invited applications from people passionate about working with technology and journalism, and keen to have an impact in this area at the BBC.

My colleague, senior product manager Andrew Leimdorfer, has this update:

We are pleased to announce that we have decided on our new Knight-Mozilla fellow, Noah Veltman, who will be starting with us in January 2013.

Noah is one of eight 2013 fellows who will all be announced at this weekend's sold-out Mozilla Festival in London who will be based in news organisations around the world, including the Guardian and the New York Times.

There are so many ways that technology is changing journalism that our first challenge is going to be to make a choice about which of these areas Noah will be helping us with next year. Working on new data visualisations and developing innovative content for mobile web will be high on the list.

We welcome Noah to the team and wish all the Knight-Mozilla fellows all the best in 2013.

Marking 15 years of the BBC online

This week marks 15 years since BBC Online was born. At about the same time, the BBC's news website also went live. The number of people visiting the news site has grown enormously over the years, and here you can see how traffic has increased, spiking at key news events, and how the appearance of the site's front page has changed over the years too. Meanwhile, for the 15th anniversary, the BBC's Director of Future Media Ralph Rivera has blogged about the significance of BBC Online today and the continuing importance of innovation to the BBC.

Expanded distribution in the US for BBC World News

This week viewers to BBC World News have been watching a series of reports focusing on the Arab uprisings, two years after they first began. Correspondents have been in Damascus, Tunis, Cairo, the Syria-Lebanon border and elsewhere. Their eyewitness TV reporting is accompanied by further explanation and analysis on our website, bbc.com/news. These are expert journalists, with years of experience and knowledge, living the story on behalf of the audience. They demonstrate our commitment to reporting the world, and bringing clarity to complex events.

Until now, however, viewers in the world's biggest TV market, the US, have found it hard to access BBC reporting of this kind. The market is saturated with TV channels, but for the past couple of years we've been very focused on securing widespread carriage on the distribution systems which bring TV into most homes.

So today the BBC is delighted to announce we have agreed to a partnership with the US cable giant - Time Warner Cable - and through this and other deals, a further 10 million homes in the US will have access to BBC World News 24 hours a day, seven days a week.

This means by the end of this year we will be available in 25 million homes, including those in most of the major markets - New York, Washington DC, Los Angeles, Chicago, Boston. There is still some way to go before we can say we have reached everyone - but 2012 has been a year of significant breakthroughs for us in the US.

The BBC is already well-known in America through its partnerships with public radio, through the success of our website BBC.com/news, and because of our nightly broadcast on public television fronted by Katty Kay. We believe our brand of high-quality, intelligent and non-partisan journalism has something to offer US audiences, and we're determined to make access to our services as simple as possible.

The timing could not be better. We're just a few weeks away from the first broadcasts of BBC World News from our brand new headquarters in central London. Three new studios, a big investment in production and journalism, and working more closely with BBC journalists working in English and 27 other languages - it's more than just a new home, it's a new start. We're delighted to share that even more widely.

BBC News comes to Burma

BBC World News will soon be available in Burma. Those are words that, even six months ago, I would not have imagined writing. But Burma, a byword for media censorship and repression, is starting to open up.

In September I visited Burma to begin the negotiations which led to this breakthrough in BBC distribution. I was struck by how rapid the media changes are for a country where state media had been long stuck in a repressive timewarp.

A World Service team visited the state broadcaster. We saw the most surreal newsroom I have ever visited. There were no journalists there. "Why not?" we asked. "We don't need them yet. The news hasn't arrived."

We learnt the news is literally delivered once a day by the state news agency. The job of the journalists was to read it out, word for word, unaltered.

But those journalists and editors are now keen to have the BBC's help in learning about open and balanced journalism. It will be a long road, given the ingrained habits of censorship and self-censorship.

But the BBC, through its pioneering media development charity BBC Media Action, is able to offer training to editors and journalists to teach them what independent journalism is. Even officials from the Ministry of Information, the former censors, asked if they could go on BBC journalism courses. Alongside the desire for training, the opening up of Burma to international broadcasters is naturally to be welcomed.

However, there is a long way to go. The massively popular BBC Burmese service, which we estimate is listened to by more than eight million people a week, is not yet allowed to broadcast within Burma. It is transmitted only on shortwave, faithfully listened to, as Aung San Suu Kyi has done for so many years. We urge the government to fully open its airwaves.

And we told the Burmese government that the BBC would continue to scrutinise the country closely. Indeed, as it becomes possible for our journalists to travel within the country, reports such as Fergal Keane's recent searing Newsnight film on human rights abuses in Rakhine state, will form a key part of the BBC's role in the country.

We will also continue to report the progress being made in the political and economic spheres.

At this early stage of opening up, it is hard to know if the hopes of media freedom will be fulfilled, but it is at least an encouraging sign that the BBC can now report from and to the country in English.

Authoritarian governments everywhere are asking themselves if they can and should hold back the free flow of news any more. And, as they ask themselves these questions, politicians, officials and journalists are looking to the BBC as the international exemplar of quality, impartial and independent journalism.

BBC World News moves to Broadcasting House

Moving BBC World News, the BBC's largest television channel, from west London to New Broadcasting House in the centre of the city is a huge project that has taken years of planning.

Relaunching and rebranding every hour of its 24 hours of output to give audiences around the world a new exciting polished HD product has made that huge project even more challenging.

Hopefully on Monday at 1200 GMT, the hard work of our 100 dedicated staff will give our audiences a bolder, brighter, more engaging look for the channel they trust to give them independent, objective news and analysis from more correspondents, in more locations, than any other international broadcaster.

Meticulous planning began about three years ago - everything from the new look of our studios to bicycle parking. We tested our studio systems - literally to breaking point - then fixed them and began the dual-running piloting that has split our newsroom teams between those keeping us on air back at Television Centre and those training and developing our programmes in our new home.

We're calling our new location The World's Newsroom because it truly reflects the world we report. We now work with colleagues from 27 language services who report for us from far flung bureaus and in London, allowing us to celebrate their unique expertise - something no other broadcaster can offer.

We'll be introducing you to those new colleagues and our new location in special live reports from inside New Broadcasting House and offering enhanced social media access so you can enjoy behind-the-scenes access.


Audiences have also told us they want to engage more with the stories we tell - to feel closer to the issues we report. We're going to help you "live the story" with us. It's our new channel ethos.

Our correspondents - expert, brave, tough, determined - live and work where they report, and we want audiences to understand their passion for the stories they cover. So expect a new style of reporting from the field. And we'll be everywhere for our relaunch with live and exclusive reports planned from Syria, China, the US and Burma to name just a few.

In the studio, trusted and familiar presenters will be sharing the day's top stories - with a sprinkling of new faces on air. We'll have a more dynamic look, with robot cameras whizzing around our studios, improved graphics and high definition screens to enhance our ability to explain and analyse. We even have some virtual reality surprises planned.

We're also developing new long-form programmes, so expect to see new hard-hitting and timely documentary series. There'll be fresh new editions of favourites such as HARDTalk with Stephen Sackur (our interrogator-in-chief), Click for the latest on tech and Health Check for medical breakthroughs.

BBC World News has come a long way since it launched as a shoestring commercial operation in a backroom at Television Centre more than 20 years ago. Our audiences have grown massively. We're required viewing from the President's White House in Washington to the President's Blue House in Seoul. And in an era when bad mortgages in the US can trigger a global economic meltdown, we know there is a huge appetite for world news delivered fast, accurately and objectively.

We hope you'll enjoy our new look. And we hope you'll join us in the world's newsroom.

BBC Arabic and the complexities of the Arab world

It is no secret that recent Arab uprisings have placed enormous burdens on the shoulders of BBC Arabic journalists responsible for reporting news from the region.

Covering the Arab world is not always an easy task - we need to mix sensible caution with a dose of courage in covering political issues that attract so many disputed views among Arabic-speaking audiences.

Our guiding principles are the BBC's values, its editorial guidelines, its ethical code, which are our reference points to maintain impartial, balanced and accurate reporting.

Across the Arab world - whether it's Tunisia, Yemen, Egypt or Libya, Bahrain, Syria and Iraq or the many other countries in the region - we know that audiences want access to objective and independent news, far removed from an agenda that favours one party, religion or sect against another. That is why audiences are turning to BBC Arabic.

Last year, our latest figures show that overall audiences to BBC Arabic have risen by more than 17% to a record high of 25.3 million adults weekly. That includes a big surge of 2.9 million in Saudi Arabia and 2.7 million in Egypt, where TV viewers in particular turned to the BBC to better understand the events happening in their own country. Our radio audiences are also holding up despite the reductions in transmission. Online is proving to be more of a challenge, but we are working hard to understand the needs of digital audiences and those for whom social media plays an increasingly important part in their lives.

In 2011, following the fall of the Mubarak leadership, we watched as ordinary Egyptians carried banners saying "Thank you, BBC!" But meeting the high expectation of audiences has a price and sometimes it's been a heavy one.

March 2011 brought a strong reminder of the risks that our staff face in covering the news - one of our reporters was arrested and tortured by Muammar Gaddafi's forces during the Libya uprising. In early 2012, our reporter in Yemen was beaten and received death threats from supporters of the outgoing president.

We are also challenged by those who disagree with our coverage. In countries such as Syria and Bahrain, BBC Arabic has been accused of bias.

The criticism comes from opposition and government alike. It may be a valid argument to say that getting criticism from both sides, in the case of Arab world certainly, is an indication of balanced coverage.

On Syria, for example, we had a series of documentaries looking at the civil war from a number of perspectives.

The first one, exploring what it's like to work for a Syrian television channel that's the mouthpiece of the government, was the butt of criticism and threats from Syrian opposition quarters. We followed it up with a programme charting a day in the lives of six Syrian women, five of whom were anti-government activists.

In our day-to-day news coverage, presenting a variety of voices from Syria is essential to us. And that is what distinguishes BBC Arabic from many media outlets in the Arab world which promote political views and agendas, and that is what we are determined to keep.

BBC Arabic marked its 75th anniversary in January. Arab politicians and ordinary people have expressed their appreciation of our track record of impartiality and trusted news. I am confident that the coming years will see further achievement on all our platforms - TV, radio and online.

School Report News Day 2013

So it's here. School Report News Day 2013 is upon us - and about 1,000 schools are due to take part, making the news that matters to them.

They will appear across BBC News - on TV, radio and online and on regional news programmes.

The project is now in its seventh year, and is bigger than ever. School reporters are in Canterbury to witness the enthronement of the new Archbishop of Canterbury Justin Welby, and we also return to the Olympic Park in London to examine the legacy from the 2012 games. And there's more - the BBC School Report website has full details of the range of topics being covered.

It is all a far cry from when we began. A small team started School Report with the aim of giving teenagers the opportunity to make the news they thought mattered. Giving them hours of BBC airtime was nerve-wracking, but it proved to be a success.

In that first year - 2007 - we worked with 12- and 13-year-olds in 120 schools. What I most remember from that year is seeing school reporters on the Six O'Clock News and thinking that this partnership between schools and the BBC had developed into something bigger than we ever thought it could be.

Fast-forward to 2013 and we are able to reach even further, both in geographical terms and into the BBC's output. We'll be broadcasting live from Radio Foyle in Londonderry, and taking over the flagship Radio 4 programme Woman's Hour.

There will also be a dedicated School Report Live channel accessible through the Red Button. We'll be updating a live news feed on our website and our @BBCSchoolReport Twitter feed throughout the day, so please follow what our school reporters are doing and let us know what you think.

iPhone and iPad app update

On Tuesday we are releasing an update to the BBC News iPhone and iPad app in the UK designed to make the app faster and more stable, with bigger, better quality images on the home screen.


We are busy doing some research and thinking at the moment about what people are looking for in our News apps in the longer term, but we thought that in the meantime, it was important to fix one or two bugs affecting some users of the existing app and to make it a better, slicker experience overall.

We want to make sure the current app remains a great way to get a quick overview of the top stories across a wide range of subjects, easy-to-scan on a mobile and, once the stories have loaded, handy to read offline too.

So, it will now be quicker to start up the app and to update it, and it should feel smoother and faster as you scroll and swipe through the screens and stories.

The larger homescreen images we've introduced serve two purposes:

  • first, you can see what's in them more clearly and there's more room for the headline
  • second, their positioning makes it clearer that you can scroll horizontally in each news category to reveal more stories (we noticed that in user testing some people assumed there were only three stories a section).

There is a new layout on iPad when you view the home screen in portrait mode - designed to show more headlines and make it easier to find the stories you're interested in.

Among the bugs that we've fixed is an issue that sometimes caused the app to get stuck when updating, and another where you sometimes saw duplicate stories within a single news category.

For our product team, these improvements required a fairly major reworking of the app's code. The good news is that they are now working from a more stable base which can be built on with new features and functionality. This revising of our code is something we've already done with our Android app, so we'll now be able to release upgrades simultaneously on both iOS and Android, which are by far the largest mobile platforms for us in terms of users. This latest update is already available internationally.

If you're a user of the app, or decide to try it out, we hope you'll like the improvements we've made. And as we think about our apps generally and plan our next steps, we'd like to hear about what you'd most like to see in future.

The Great British class calculator

We've had a huge response to our class calculator this week, particularly across social media, following a major survey by BBC Lab UK. The survey suggests that traditional categories of working, middle and upper class are outdated and we all fit in to one of seven new classes.

The class calculator - which lets you work out where you might fit in amongst the new categories - has attracted about six million page views on the BBC News site, making it the second most popular article of 2013 to date. (The most viewed article this year has been the helicopter crash in Vauxhall in January.) Nearly 1.9 million of those views have come from those of you accessing the site on mobiles and tablets.

But one thing that really stands out is how widely the story has been shared across social media, with more than 300,000 shares so far. More than a quarter of links to the calculator have come from social networking sites.

More than half a million referrals came from Facebook alone, and about 107,000 from Twitter. This is a much higher number than we usually see shared across social media. If you compare the class calculator with the other top stories of the week, usually about 5% of known referrals come from social media sites.

So why has it proven so popular with our audience? Michael Orwell, a producer at BBC Lab UK, worked closely on the survey and said one of the best things about the project was that the audience contributed to new research with top academics.

The calculator itself, produced by the BBC News Visual Journalism team in collaboration with BBC Knowledge and Learning, lets everyone engage with the new model and discover where they might fit in.
